沿皮浅刺内关公孙穴对脑梗塞大鼠Bcl-2 Bax的影响

摘    要:目的:研究浅刺针法对脑缺血大鼠细胞凋亡调控基因Bcl-2(B cell lymphoma/leukemia-2)、Bax(Bcl-2Associated X protein)的影响。方法:将240只Wistar大鼠随机分造模组和空白组,采用血栓栓塞法建立脑梗塞大鼠模型,后将造模成功的180只大鼠随机将为浅刺针法组(Ⅰ组)、常规针刺组(Ⅱ组)、模型对照组(Ⅲ组),空白组为正常对照组(Ⅳ组)。通过神经体征评分、脑组织Bcl-2和Bax蛋白的表达等几个指标,观察经过治疗后的Ⅰ、Ⅱ组大鼠和相应时间未经治疗的III、IV组大鼠的大脑细胞凋亡情况。结果:3个时间点的浅刺针法组大鼠脑组织Bax蛋白表达明显低于模型组和常规组针刺组,而Bcl-2则明显高于模型组和常规组针刺组(P<0.01),说明浅刺针法可有效改善脑梗塞大鼠的神经缺损体征。结论:浅刺内关、公孙穴位通过下调Bax蛋白表达和上调Bcl-2蛋白的表达,抑制细胞凋亡,发挥神经保护作用。并在分子水平证实经络在体表有其物质基础,浅刺可取得有效的刺激作用,使浅部组织的经络特异性能充分显示出来。

Abstract:Objective:To study shallow Spike Act of rat brain after cerebral infarction is chemic penumbra of Bcl-2,Bax effect.Methods:120 Wistar rats were randomly divided into shallow Spike Act(Ⅰ),conventional acupuncture group(Ⅱ),the model group(Ⅲ),and normal control group (Ⅳ).Thromboembolism three groups using the rat model of the establishment of cerebral infarction.Ⅳ group without any treatment,but at the same time bundled fixed.Then each group were randomly divided into three days after 2,7,14 point in time,each of the five time points in rats.Neural signs from the score,using immunohistochemical methods of brain tissue Fas,Bcl-2 protein expression changes observed after 2,7,14 days after treatment Ⅰ,Ⅱ rats and the corresponding time untreated III,IV rat brain cell apoptosis.Results:Three time points shallow Spike Act rat brain ischemic penumbra fas protein expression was significantly lower than the model group and the conventional group acupuncture group,and bcl-2 was significantly higher than that model and the conventional group needle thorn group(P〈0.01),indicating that shallow Spike method can be effectively improved cerebral infarction in rats nerve defect signs.Conclusion:Bax,Bcl-2 in rat brain ischemic cerebral infarction cell apoptosis plays an important role,thorn shallow ditch,which customs very neiguan,gongsun point reduction through increased and expression of Bax protein Bcl-2 protein expression,inhibition of apoptosis,a neuroprotective effect.And Meridian confirmed at the molecular level in the body has its material foundation,shallow thorns can be an effective stimulus to shallow Meridian specific organizations can fully displayed.
