抗纤灵颗粒对单侧输尿管梗阻模型大鼠肾组织NO cNOS iNOS的影响

摘    要:目的:观察抗纤灵颗粒对单侧输尿管梗阻(UUO)模型大鼠肾组织一氧化氮(NO)、结构型一氧化氮合酶(cNOS)、诱导型一氧化氮合酶(iNOS)的影响。方法:将SD大鼠随机分为正常组、假手术组、模型组、抗纤灵组、氯沙坦组。造模后7、14、21、28天分批处死,进行肾组织匀浆,检测NO及cNOS、iNOS的含量。结果:模型组大鼠NO含量明显降低、cNOS活力显著下降,iNOS活力显著升高,与正常组及假手术组比较有统计学意义(P<0.01);抗纤灵组大鼠NO含量显著升高、cNOS活力明显增强,iNOS活力明显下降,与模型组比较有统计学意义(P<0.01,P<0.05),与氯沙坦组比较,有统计学意义(P<0.01,P<0.05)。结论:抗纤灵颗粒通过上调cNOS诱导的NO含量,下调iNOS诱导的NO含量,引起TGF-β1表达下调,减轻肾间质纤维化,延缓肾小球硬化,从而对肾脏起到一定的保护作用。

Abstract:Objective:To observe the effect of Kangxianling on NO,cNOS and iNOS in renal tissue in rats with unilateral ureteral obstruction.Methods:SD rats were randomly divided into normal group,sham-operation group,model group,Kang xianling(KXL) group and Losartan group.After making model for 7,14,21,28d.The animals were killed in batch to get their kidney tissue to determine the levels of NO,cNOS and iNOS in renal homogenate.Results:The concentration of NO,cNOS in model group decreased obviously,and that iNOS increased instead,which were significantly different with normal group and sham-operation group(P〈0.01).The concentration of NO,cNOS in KXL group increased obviously,and that iNOS decreased instead,which were significantly different with model group and Losartan group(P〈0.01,P〈0.05).Conclusion:Through enhancing NO induced by cNOS and declining NO induced by iNOS,KXL can decrease the expression of TGF-beta 1,expand vessel of kidney,lighten and slow down the fibrosis of renal interstitium and renal glomerulus,so to has some effect of protecting the kidney.
