促红细胞生成素对新生大鼠缺氧缺血脑损伤前列腺素代谢的影响

摘    要:目的:探讨促红细胞生成素(erythropoietin, EPO)对缺氧缺血新生大鼠脑组织血栓素B2(TXB2)及6-酮-前列腺素F1ɑ(6-K-PGF1ɑ)含量的影响。方法:选用7日龄wistar大鼠80只,制备缺氧缺血性脑损伤(HIBD)模型后注射促红细胞生成素(EPO)及丹参注射液。缺氧缺血48 h取脑组织,用放射免疫分析法测量新生大鼠脑组织中TXB2、6-K-PGF1ɑ含量变化。结果:缺氧缺血模型组与生理盐水对照组TXB2与6-K-PGF1ɑ显著增高,与正常组比较有显著性差异(P<0.01)。EPO组与丹参组6-K-PGF1ɑ低于模型组及生理盐水对照组,有显著性差异(P<0.01), TXB2差异不明显。丹参组与EPO组之间TXB2与6-K-PGF1α无显著性差异(P>0.05)。结论: EPO通过预防和减轻TX及PG代谢紊乱对缺氧缺血新生大鼠脑损伤起保护作用。

Effects of erythropoietin on prostaglandin metabolism of newborn rats with cerebro cellular damage of hypoxic-ischemic
Abstract:Objective:To study the effects of erythropoietin(EPO)on the TXB2and 6-K-PGF1ɑ level in brain tissue of hypoxic-ischemic neonatal rats.Methods:80 numbers of 7-day old newborn Wistar rats were selected to establish HIBD model,and were injected EPO and saliva militimbixae injection,then the rats were killed after hypoxia and ischemia for another 48 hours.And detect TXB2 and 6-K-PGF1ɑ levels in brain tissue using RIA.Results:TXB2 and 6-K-PGF1ɑ level in brain of HIBD groups and saline group were higher than that of the normal group,there were significant different(P<0.01),6-K-PGF1ɑ level of EPO group and saliva militimbixae group were much lower,they were significantly different from that of HIBD group and normal saline group(P<0.01),so as to TXB2.But there's no significant of TXB2 and 6-K-PGF1ɑ level between saliva and EPO group.Conclusion:EPO can protect brain of HIE newborn rats from injury through prevent and deduce the disorder of TX and PG metabolism.
