清除致癌物多环芳烃的中药组分复方配伍研究

摘    要:目的:基于均匀设计方法研究多种中药有效组分体外清除致癌物多环芳烃的最佳配伍作用.方法:采用改进最小角(least angle regression,LABS)回归分析,通过荧光光谱法进行甘草总黄酮、银杏叶提取物、淫羊藿总黄酮和黄芪总苷4个中药组分不同配比清除多环芳烃的作用研究.结果:通过LRAS回归分析获得了中药组分配伍清除多环芳烃的回归方程,优化组合后进行药效学验证,结果显示该中药组分配伍最佳剂量组合为甘草总黄酮3.144 g·L-1、银杏叶提取物0.8g·L-1、淫羊藿总黄酮0.024 g·L-1和黄芪总苷0.036 g·L-1.结论:上述中药组分配伍后能有效清除致癌有害物质多环芳烃;本实验采用的均匀设计结合荧光光谱检测方法,为筛选评价中药组分配伍清除致癌物多环芳烃的作用提供了合适的研究方法.

Abstract:Objective: To find a suitable way of quantitative composition-activity relationship of Chinese herbal for reducing the carcinogen of polycyclic aromatic hydrocarbons(PAHs) based on uniform design. Method: Chinese herbal active ingredient formula was composed of total flavonoids of glycyrrhiza (TFG), ginkgo biloba extract (GBE), total flavonoids of epimedium (TFE) and astragaloside (AST). The herbal efficacy was detected by scavenging polycyclic aromatic hydrocarbons in vivo. And the mathematical model was constructed to describe the connection between different combinations and their scavenging capacity by the method of least angle regression(LARS) regression. Result: The best effect was in the ratio of TFG 3.144 g·L-1,GBE 0.8 g·L-1,TFE 0.024 g·L-1, AST 0.036 g·L-1. Conclusion: Uniform designed research is one of efficient ways to describe the quantitative composition-activity relationship for reducing the carcinogen of PAHs.
