降糖防聋方对体外高糖培养成骨细胞骨钙素分泌的影响

摘    要:目的:探讨降糖防聋方对体外高糖培养成骨细胞骨钙素的影响.方法:采用改良的组织块法分离培养新生大鼠颅骨成骨细胞,用不同浓度的降糖防聋方含药血清及不含降糖防聋方含药血清的α-MEM培养液加入到成骨细胞高糖(糖终浓度为300 kg·L-1)培养体系,作用不同时间,应用放免法测定细胞骨钙素(BGP)的水平.结果:100,50,10 mg·L-1的降糖防聋方含药血清对体外高糖培养成骨细胞具有明显的促进作用(P<0.05),在较高浓度对对成骨细胞增殖无明显抑制效应.结论:降糖防聋方含药血清对高糖培养成骨细胞骨钙素有不同程度调节改善作用.

Abstract:Objective:To study the Jiangtang Fanglong fang on high glucose cultured osteoblast osteocalcin secretion in vitro.Method:Modified separation of tissue was used to culture neonatal rat calvaria osteoblasts.The α-MEM medium with different concentrations of serum containing Jiangtang Fanglong fang and drug-free serum were added to the osteoblasts cultured in high glucose system(final concentration of sugar).The levels of osteocalcin were measured by radioimmunoassay.Result: The serum containing Jiangtang Fanglong fang with 100,50,10 mg·L-1 had a significant promoting action.The higher concentrations had no significant inhibitory effect on the proliferation of osteoblasts.Conclusion:The serum containing Jiangtang Fanglong fang can regulate in vitro cultured osteoblast osteocalcinsecretion.
