miR-139-3p在低氧诱导凋亡的心肌细胞中的表达及其作用研究

摘    要:目的:探讨微小RNA-139-3p(miR-139-3p)在低氧诱导的原代心肌细胞凋亡模型中的表达及其意义。方法:在正常和低氧条件下,采用RT-qPCR检测乳大鼠原代心肌细胞miR-139-3p的表达水平;进一步将miR-139-3p抑制剂和miR-139-3p抑制剂阴性对照转染到心肌细胞后,将细胞置于37℃密闭的缺氧盒中(95%N_2和5%CO_2)培养12 h,采用流式细胞术和Western blot法检测细胞凋亡情况。结果:低氧培养12 h后,与正常培养组(n=3)比较,低氧组(n=3)心肌细胞中的miR-139-3p相对表达量显著上调(P<0.05)。低氧组心肌细胞凋亡率也显著升高(P<0.05)。与miR-139-3p抑制剂阴性对照组(n=3)比较,miR-139-3p抑制剂组(n=3)的心肌细胞凋亡率显著降低(P<0.05)。结论:miR-139-3p在低氧诱导的原代心肌细胞凋亡模型中表达上调。抑制miR-139-3p的表达能降低低氧诱导的心肌细胞凋亡。

Abstract:AIM:To investigate the expression and clinical significance of microRNA-139-3p (miR-139-3p) in the apoptosis model of cardiomyocytes induced by hypoxia. METHODS:Under normal and hypoxic conditions, the expression of miR-139-3p in neonatal rat cardiomyocytes was detected by RT-qPCR. miR-139-3p inhibitor and miR-139-3p inhibitor negative control were transfected into the primary neonatal rat cardiomyocytes. The transfected cardiomyocytes were cultured in closed anoxic box (95% N2 and 5% CO2) at 37℃ for 12 h. Flow cytometry and Western blot were used to determine the apoptosis of cardiomyocytes. RESULTS:After hypoxia for 12 h, the expression level of miR-139-3p and the apoptotic rate of the cardiomyocytes in hypoxia group were significantly higher than those in normal group (P<0.05). Moreover, compared with the miR-139-3p inhibitor negative control group, the apoptotic rate of the cardiomyocytes was significantly decreased in miR-139-3p inhibitor group (P<0.05). CONCLUSION:The expression of miR-139-3p is signi-ficantly increased in apoptotic neonatal rat cardiomyocytes induced by hypoxia. Inhibition of miR-139-3p expression reduces hypoxia-induced apoptosis of cardiomyocytes.
