NF-κBp65在肝癌血管生成中的作用

摘    要:目的本实验通过对肝细胞癌(HCC)组织、癌旁组织、肝硬化组织及正常肝组织中NF-κBp65蛋白表达和微血管密度(MVD)进行检测,探讨NF-κBp65的蛋白表达与HCC新生血管生成的关系,初步阐述NF-κBp65在HCC血管生成中的机制。方法选用45例肝细胞癌组织和32例癌旁肝组织、15例肝硬化组织、10例正常肝组织标本作对照,采用免疫组化法(SABC)检测NF-κBp65的表达和MVD值,并结合患者的临床病理指标进行统计学分析。结果NF-κBp65在HCC中的表达及MVD值显著高于癌旁肝组织、肝硬化组织和正常肝组织(P<0.05);HCC组织中NF-κBp65阳性表达组显著高于阴性表达组(P<0.05);血管侵犯组显著高于无血管侵犯组(P<0.05)。结论NF-κBp65在HCC组织中的表达高于癌旁肝组织、肝硬化组织和正常肝组织;HCC中NF-κBp65的表达与肝癌血管生成有关。

Abstract:Objective By testing the expression of NF-κBp65and by counting micro vascular density(MVD) in HCC tissues, Para tumor tissues, cirrhosis liver tissues and normal liver tissues, to determine the relationship between the former three factors and angiogenesis in HCC tissues, furthermore to document the mechanism of NF-κBp65 in angiogenesis of HCC. Methods Forty-five tumor tissue samples and thirty-two tumor-surrounding tissues samples were obtained from primary HCC patients undergoing surgical resection. Fifteen cirrhosis liver tissues and ten normal liver tissues samples were taken from previously patients with a benign tumor. SABC method of Immunohistochemistry was used to determine the relationship between their expression and clinicopathological parameters.Results Statistical analysis revealed that NF-κBp65-positive stain rate was higher in tumor tissues than that in tumor-surrounding tissues, cirrhosis liver tissues and normal liver tissues. (P<0.05). Numerical value of MVD was significantly higher in tumor tissues than that in tumor-surrounding tissue, cirrhosis liver tissue and normal liver tissue (P<0.05). Numerical value of MVD was higher in tumor tissue samples with NF-κBp65 positive stain than that in their negative tumor tissue samples. (P<0.05). The expression of NF-κBp65 in vascular invasion of tumor tissue samples was higher than that in vascular non invasion of tumor tissue samples. (P<0.05) .Conclusion NF-κBp65 positive stain rate were higher in the tumor tissue than that in tumor-surrounding tissue, in cirrhosis liver tissue and in normal liver tissue. The over expression of NF-κBp65 in the hepatocellular carcinoma were associated with tumor angiogenesis of HCC.
