胰腺炎时胰腺细胞反应性的变化及栀子的影响

摘    要:取SD大鼠30只(200~300g)随机分为3组。20只动物用1.5%DCA-Na经胰管逆行注射诱发急性胰腺炎,其中半数于术前1小时,术后4小时以及细胞学实验前1小时经口给50%栀子煎剂各2ml,另一半给生理盐水,处理相同。术后24小时全部动物及10只未作处理的正常动物被处死,用肢原酶制备胰腺细胞悬液。在给CCK和氨甲酰胆硷前后测上清液中的淀粉酶检测细胞的反应性。实验揭示胰腺炎动物基础状态下淀粉酶的释放显著高于正常,但在CCK或氨甲酰胆硷刺激下的释放量则明显减少,然栀子组在刺激前后的释放量均显著地更少。结果表明基础状态下淀粉酶释放量的增多系炎症引起胰腺腺泡障害所致,而对刺激因子表现的反应性释放减少则可能是胰腺炎时的一种自身适应性保护机制,同时也表明栀子煎剂具有减轻这种障害和增强保护机制的作用。

Abstract:30 SD rats weight 200-300g were randomly divided into 3 groups. 20animals were induced acute pancreatitis with 1.5% DCA-Na throughintra-pancreatic duct retrograde injection. Among which half of themwas treated with 50% GJE decoction 2ml per oral 1h preand 4h post-operation and also at 1h before cellular examination, another half wastreated as same but with saline. 24h after operation all of them andalso other 10 untreated rats as normal control were sacrified, and thepancreatic cellular suspension was prepared with collangase. Thecontent of amylase in supernatant would be detected before and aftertreated with CCK and carbacol as cellular responsibility measurement.It was demonstrated that the release of amylase in supernatant atbasic condition was significantly higher, but less released after CCKor carbacol stimulation in pancreatitis groups than the normal,mean while, the content of amylase either before or after stimulationwas all lesser in the GJE group. It was seemed that the raise of amy-lase release in basic condition may be a disorder of pancreatic acini caused by inflammation and the decrease of responsive release of amy-lase to stimulators may be a self-adaptive protection mechanism inpancreatitis, and it also showed that GJE decoction would reduce thedisorder and enhance this protective mechanism.
