白细胞介素-1α对培养的人眼小梁细胞吞噬功能的影响

摘    要:目的观察白细胞介素-1α(IL-1α)对体外培养的人眼小梁细胞(HTM)吞噬功能的影响。方法取人眼小梁组织进行小梁细胞体外培养,取传3~5代的人眼小梁细胞,加入不同浓度的IL-1α的DMEM-F12培养液,用倒置相差显微镜、光镜、电镜观察细胞吞噬功能的变化。结果小梁细胞吞噬乳胶微球数与IL-1α浓度(F=3.603,P=0.046)和作用时间(F=15.846,P=0.005)均有相关性(F=18.069,P=0.000)。结论IL-1α可以直接损害体外培养的人眼小梁细胞吞噬功能,其吞噬程度与IL-1α作用时间和浓度相关。

Abstract:OBJECTIVE: To investigate the effect of interleukin-1alpha (IL-1alpha) on the phagocytic function in cultured human trabecular meshwork (HTM) cells. METHODS: HTM Cells were cultured and subcultured in vitro. The third to fifth passage of HTM cells were treated with different concentrations of IL-1alpha or vehicle in DMEM/F12 medium. 1000 of 0.85 microm microsphere/per cell were added to each well. The morphological changes of HTM cells were observed by an inverted phase-contrast microscope and electron microscope. The cellular phagocytic function of HTM cells was evaluated by counting the numbers of microsphere in 20 HTM cells using a light microscope. RESULTS: The break of HTM cell membrane was found under the electron microscope. The numbers of microsphere in HTM cells was significantly (P = 0.005) reduced in a dose- and incubation time-dependent manner (F = 3.603, P = 0.046, F = 15.846, P = 0.005, respectively). CONCLUSIONS: IL-1alpha affects the phagocytic function of HTM cells that is correlated to the concentration and the incubation time.
