肺癌患者荷瘤DNA倍体含量及外周血FOXP3与化疗效果的相关性研究

摘    要:目的探讨肺癌患者荷瘤DNA倍体含量及外周血FOXP3与化疗效果的相关性。方法采用流式细胞术检测89例肺癌患者的DNA倍体含量,并对67例手术和化疗患者检测体内CD4+CD25+FOXP3+调节性T淋巴细胞(Treg细胞),并以63例健康体检者作正常对照。结果随着TNM分期的发展,异倍体肿瘤的发生率逐渐上升(P0.01),有淋巴结转移或癌栓者其癌旁肺组织异倍体比率高于无癌栓或淋巴结转移者,肺癌患者CD4+CD25+T细胞、CD4+FOXP3+T细胞占CD4+T细胞的比例与对照组相比明显升高,CD4+FOXP3+T细胞占CD4+CD25+T细胞的比例与对照组相比无差异,化疗后较化疗前显著降低。结论化疗可使患者CD4+FOXP3+T细胞占CD4+及CD4+CD25+T细胞的比例明显下调,同时测定癌及癌旁细胞核DNA倍体模式,在评估肺癌恶性程度及预后上有一定意义。

Abstract:Objective To study the lung cancer patients with tumor DNA ploidy contem and effect of peripheral blood FOXP3 associated with chemotherapy. Methods To detect the DNA ploidy content of 89 eases lung cancer patients by flow cytometry and to detect 67 patients with surgery and diemotherapy of CD4^+ CD25^+ FOXP3^+ regulatory T cells (Treg cells), and 63 cases of healthyindividuals as normal control. Results as the development of TNM staging aneuploid tumors gradually increased in the incidence( P 〈 0.01), with lymph node metastasis or tumor thrombus by its adjacent lung tissue aneuploidy rate Higher than in those without lymph node metastasis or tumor thrombus, CD4^+ CD25^+ T cells, CD4^+ FOXP3 ^+T cells of lung cancer patients higher compared with the control group, CD^+ FOXP3^+T cells accounted for CD4^+ CD25^+ T cells in the proportion of no significant difference compared with the control group, After chemotherapy compared with chemotherapy were significantly lower. Conclusion Chemotherapy allows the proportion of patients with CD4^+FOXP3^+ T cells accounted for CD4+ and CD4^+ CD25^+ T cells significantly reduced, Simultaneous deter-ruination of cancer and adjacent nuclear DNA ploidy pattern, in assessing the degree of malignancy and prognosis of lung cancer, there is a certain sense.
