埃博霉素异源表达研究进展

摘    要:埃博霉素是继紫杉醇之后新一代的微管蛋白抑制剂,被认为是目前最有希望的抗癌药物之一。但是,目前发酵产量较低,化学合成步骤多,不能实现工业化生产。相比较而言,埃博霉素的异源表达有很多优点,比如异源宿主代时较短,易于基因工程操作等。在异源宿主中产生埃博霉素已经成为目前研究的一个热点。本文综述了近年来埃博霉素在天蓝色链霉菌(S.coelicolor),大肠杆菌(E.coli)和橙黄色黏球菌(M.xanthus)等宿主中表达的研究进展以及面临的机遇和挑战。

Abstract:The polyketide natural products epothilones are promising anticancer agents that stabilize microtubules as paclitaxel(Taxol) does. However, epothilones cannot be mass-produced because of their lower output in fermentation and complicated procedures in chemical synthesis. The production of epothilones in heterologous hosts offers many advantages over the use of the natural host, including easy genetic manipulation and quick replication of heterologous hosts. Production of epothilones in heterologous hosts has become a hot spot. This review briefly summarizes heterologous production of epothilones in S. coelicolor, E. coli and M. xanthus.
