疲劳型亚健康小鼠模型的研制

摘    要:目的 研制疲劳型亚健康小鼠模型.方法 疲劳型亚健康小鼠模型制备:强迫小鼠水中站立8 h/d,分别观察连续造模5、9、12、16 d时小鼠行为学表现,以力竭游泳时间反映其疲劳程度,结合血浆生化、血常规及病理组织学检查等,确定造成疲劳型亚健康的最佳时间.疲劳型亚健康小鼠模型的评价:对上述创建的疲劳型亚健康小鼠模型进行评价,观察模型小鼠的疲劳持续时间.结果 疲劳型亚健康小鼠模型制备:模型各组游泳时间与对照组比较显著下降(P=0.000).与对照组小鼠比较,造模16 d组BUN、ALT、AST、TG改变明显(P<0.05);其余血生化、血常规及病理学检查未见明显异常.疲劳型亚健康小鼠模型的评价:模型小鼠力竭游泳时间明显下降;去除造模因素,休息恢复7 d后其力竭游泳时间有增长,与对照组比较差异有统计学意义(P<0.05);至恢复14 d后,小鼠力竭游泳时间与对照组比较差异无统计学意义(P>0.05).结论 采用强迫小鼠水中站立8 h/d,连续9 d,成功制备疲劳型亚健康小鼠模型.该模型的稳定性、重复性较好,为疲劳型亚健康病理机制的研究及相关药物的筛选提供技术平台.

Abstract:Objective To establish and evaluate the fatigue-type sub-health mouse model. Methods Experiment 1: Mice in model groups were forced to stand in water 8 hours per day each for 5,9,12 and 16 days.The performances of mice behaviors,degree of fatigue,pathological signs in tissue,blood biochemistry results and other indicators were analyzed to determine the optimal time for this fatigue-type sub-health model.Experiment 2: Evaluation of the model was performed with the test of 7% weight-loaded forced swim.Results Experiment 1: In comparison with control group,the exhaustive swimming time was significantly shortened in all model groups(P=0.000).Significant alterations of BUN,ALT,AST and TG were only observed in model 16d group(P<0.05),no significant change was revealed in the other model groups.Experiment 2: The exhaustive swimming time was significantly reduced in model mice.Although the fatigue degree abated after a rest of 7days free of modeling factors,swimming time was still significantly shorter than normal level(P<0.05).However,the swimming time became no significant difference compared with that of normal control mice with a recovery of 14 days(P>0.05). Conclusion Forcing mice to stand in water 8 hours per day for 9 days can successfully establish this mouse model of fatigue-type sub-health,with significant stability and reproducibility,providing foundation for further study of fatigue-type sub-health.
