摘    要:用化学发光法检测粒细胞吞噬功能具有灵敏、准确、快速和用样品量少等优点。在加入 luminol 增强发光后,用发光仪监测,灵敏度可以达到用液闪计数器测量的同等水平(粒细胞悬液浓度10~5/ml 或更低些),测定样品体积可降到1ml 左右,而且其突出的优点是可以在生理温度(37℃)下恒温测定,操作方便,不必作避光处理,既可应用于粒细胞吞噬功能的机理研究,也可应用于临床监测,对烧伤病人粒细胞吞噬功能的化学发光测定所得初步结果,显示了这一方法的应用潜力。


STUDIES ON THE RELATIONSHIP BETWEEN THE PHAGOCYTOSIS OF HUMAN GRANULOCYTES AND THE REACTIVE OXYGEN SPECIES(ROS)I.PHAGOCYTOSIS DETERMINATION BY CHEMILUMINESCENCE(CL)MEASURMENTS
Abstract:The phagocytosis by human polymorphonuclear leukocytes(PMNs)has been monitor- ed by measuring the chemiluminescence(CL)accompaning the stimulation by opsonized particles(bacteria or zymosan).The parallel relationship between phagocytosis and CL has been demonstrated.The CL has been enhanced by adding luminol and can be measured on a LKB Luminometer 1250.This luminol-amplified assay system have shown some advantages compared with other phagocytic assays:(a)sensitivity and reproduci- ablity requiring,less than 10~5/ml PMNs and the reduction of total assay volume to 1 ml. (b)easy to operate and rapidness,avoiding the necessity of working under subdued red light.(c)physiologic condition,the possibility or continuous measurment under physiologic temperature(e.g 37℃).It is concluded that this assay system is suitable for mechanism studies of phagocytosis as well as for clinic monitoring.
