正交试验优选天麻安神颗粒乙醇提取工艺

摘    要:目的:优选天麻安神颗粒的乙醇提取工艺。方法:以天麻素、五味子醇甲、钩藤碱总提取量及出膏率为综合评价指标,通过L9(34)正交试验考察乙醇体积分数、乙醇用量、提取时间及提取次数对天麻安神颗粒醇提工艺的影响。采用HPLC测定3种指标成分的含量,天麻素流动相乙腈-0.05%磷酸(3∶97),检测波长220 nm;五味子醇甲和钩藤碱流动相乙腈-0.001%三乙胺(45∶55),检测波长250 nm。结果:最佳提取工艺为加6倍量50%乙醇提取3次,每次1 h。天麻素、五味子醇甲、钩藤碱提取量分别为2.69,1.47,0.18 mg·g-1,干膏率22.64%。结论:优选的醇提工艺稳定可行,适用于天麻安神颗粒的工业化生产。

Abstract:Objective: To optimize extraction technology of Tianma Anshen granules. Method: With dry extract rate and contents of gastrodin, schizandrin, rhynchophylline as comprehensive index, orthogonal design was adopted to investigate effects of the amount and concentration of ethanol, extracting time and times on extraction technology.Contents of gastrodin, schizandrin and rhynchophylline were determined by HPLC, mobile phase of gastrodin was acetonitrile-0.05% phosphoric acid (3:97), detection wavelength was 220 nm;mobile phase of schizandrin and rhynchophylline was acetonitrile-0.001% triethylamine (45:55), detection wavelength was 250 nm. Result: Optimal extraction technology was extracted 3 times with 6 times the amount of 50% ethanol for 1 h per time.Extraction amounts of gastrodin, schizandrin and rhynchophylline were 2.69, 1.47, 0.18 mg·g-1, dry extract rate was 22.64%. Conclusion: This optimized extraction technology is stable, feasible and suitable for industrial production of Tianma Anshen granules.
