摘    要:目的:探讨杨桃根总提取物(YTG)对链脲佐菌素(STZ)诱导的糖尿病心肌病大鼠心脏功能的影响。方法:雄性Wistar大鼠给予高糖高脂饮食4周后,用负荷小剂量的STZ ip建立糖尿病大鼠模型。将造模成功的大鼠随机分为模型组,二甲双胍组,YTG高、中、低剂量组(0.6,0.3,0.15 g·kg-1·d-1),另设正常组,每组8只,ig给药4周,分别测定空腹血糖(FBG),体重,甘油三酯(TG),低密度脂蛋白(LDLC),总胆固醇(TC),肌酸激酶(CK),天冬氨酸转氨酶(AST),肌酸激酶同工酶(CKMB),乳酸脱氢酶(LDH),α-羟丁酸脱氢酶(HBDH),丙二醛(MDA)的含量以及总超氧化物歧化酶(T-SOD)的活性,并进行心肌组织HE染色。结果:与正常组比较,模型组FBG,TG,LDLC,TC,CK,AST,CK-MB,LDH,HBDH,MDA含量明显升高,体重明显降低,T-SOD的活性明显降低(P0.05);与模型组比较,各YTG治疗组FBG,TG,LDLC,TC,CK,AST,CK-MB,LDH,HBDH,MDA含量降低,体重明显升高,T-SOD的活性升高(P0.05)。与正常组比较,模型组心肌细胞排列紊乱,细胞肥大变形,细胞核深染,间质纤维化明显;与模型组比较,各给药组心肌细胞肥大变形有所改善,间质纤维化得到明显改善。结论:YTG可减轻糖尿病心肌病大鼠心肌功能的损害,对心脏功能有一定的改善作用,其机制可能与清除自由基有关。

Abstract:Objective: To examine the potential effect of Averrhoa carambola root (Yaotaogen, YTG) extract on streptozotocin (STZ)-induced diabetic cardiomyopathy in rats. Method: Male Wistar rats fed a high glucose and hyperlipid diet for 4 weeks. Then diabetes mellitus model was induced by a single injection of STZ. Diabetes mellitus rats were divided into the metformin group, the model group, the high-, medium-, low-dose YTG extract groups (0.6, 0.3, 0.15 g·kg-1·d-1). Another 8 rats were assigned to the normal group. After 4 weeks, the levels of fasting blood-glucose (FBG), body weight (BW), triglyceride (TG), low density lipoprotein (LDLC), total cholesterol (TC), creatine kinase (CK), aspartate aminotransferase (AST), creatine kinase isoenzyme (CK-MB), lactic dehydrogenase (LDH), hydroxybutyrate dehydrogenase (HBDH), malondialdehyde (MDA), and the vitality of total superoxide dismutase (T-SOD) were measured. The hematoxylin-eosin (HE) staining of myocardial tissue was conducted. Result: Compared to the normal group, the levels of FBG, CK, AST, CK-MB, LDH, HBDH, and MDA increased, BW and the vitality of T-SOD decreased in the model group (P<0.05). Compared to the model group, the levels of FBG, TG, LDLC, TC, CK, AST, CK-MB, LDH, HBDH and MDA decreased, BW and the vitality of T-SOD increased in all doses of YTG groups (P<0.05). Moreover, YTG could improve cell hypertrophy deformation and interstitial fibrosis apparently. Conclusion: YTG extract has a beneficial effect on heart function by relieving the heart damage in diabetic cardiomyopathy. The mechanism may be related to removing the free radicals.
