摘    要:目的:明确延寿液对大鼠吸入臭氧所致皮肤衰老的延缓作用,探讨其作用机制。方法:雄性SD大鼠随机分为6组,分别为正常组,模型组,维生素E组(13.5 mg·kg-1),延寿液低、中、高剂量组(45,90,180 mg·kg-1),每组8只,正常组吸入空气,造模组吸入臭氧(1.2±0.2)mg·L-110 h·d-1×45 d染毒,造模的同时给予延寿液。45 d后动脉取血获得血清与血浆,取大鼠两肩胛至两髂骨之间的背部皮肤,检测血浆谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶(CAT)活性,血清超氧化物歧化酶(SOD)活性和过氧化脂质(LPO)含量,皮肤羟脯氨酸(Hyp)含量,皮肤组织学观察。形态计量表皮厚度、弹性纤维、胶原纤维、网状纤维、肥大细胞颗粒的体积比,比较组间变化的显著性。结果:与正常组比较,模型大鼠GSH-Px,CAT,SOD活力明显下降(P0.01),LPO含量明显升高(P0.01),Hyp含量明显降低(P0.01);表皮变薄,基底细胞减少,胶原纤维排列稀疏,弹性纤维变短,数量减少,网状纤维较少,出现肥大细胞脱颗粒。延寿液低、中、高剂量组均能明显升高GSH-Px,CAT,SOD活力(P0.05,P0.01),明显降低LPO含量(P0.01),明显升高Hyp含量(P0.05,P0.01),胶原纤维、弹性纤维和网状纤维均增多,增加表皮厚度,减少肥大细胞脱颗粒现象。结论:通过抑制炎症反应、恢复组织结构,延寿液可延缓大鼠吸入臭氧诱导的皮肤衰老。

Abstract:Objective: To obverse the effect of Yanshou decoction on skin aging in rats induceded by inhaling ozone and to explore its mechanism. Method: Fourty-eight male SD rats were randomly divided into six groups:the normal group, the model group, the Vitamin E group (13.5 mg·kg-1), the low-, medium-, and high-dose Yanshou decoction groups (45, 90, 180 mg·kg-1), 8 in each group. Rats in the normal groups were exposed to fresh air. The other rats were exposed to ozone (1.2±0.2) mg·L-1 4 hours daily for consecutive 45 days. Meanwhile, the corresponding medicines were intragastrically administrated once daily for consecutive 45 days. The activity of serum superoxide dismutase (SOD), plasma glutathion peroxidase (GSH-Px) or plasma catalase (CAT), the levels of serum lipid peroxidase (LPO) or dermis hydroxyproline (HYP) were detected and the cutaneous histopathology was observed. Morphometry of skin including epidermis thickness, collagen fiber, elastic fiber and reticular fiber ratio of changes in rats were compared. Result: Compared with the normal group, the activity of SOD, the contents of LPO in serum decreased, the activity of GSH-Px and CAT in plasma decreased, the contents of Hyp in skin tissue homogenate decreased in the model group (P<0.01). Moreover, it was discovered that the epidermis was thin, the basal cells decreased, few collagenous fiber was sparsely arranged, elastic fiber was shorter, and mast cell degranulation were present. Compared with the model group, the activity of SOD, the contents of LPO in serum and the activity of GSH-Px, CAT in plasma increased, the contents of Hyp in skin tissue homogenate increased in all doses of Yanshou decoction groups (P<0.01, P<0.05). The contents of soluble collagen, collagen fiber, elastic fiber and reticular fiber increased and the epidermis was thicker in the Yanshou decoction groups. Moreover, Yanshou decoction could prevent the skin mast cells of rats from releasing granules induced by ozone. Conclusion: Yanshou decoction could postpone the process of skin aging in rats inhaling ozone.
