微波辅助提取补阳还五汤中多糖的含量及测定

摘    要:目的提取补阳还五汤中的多糖并测定其含量。方法用微波辅助法提取,料液比为1∶7,提取时间20min,微波功率325w。用蒽酮比色法测定多糖的含量,加蒽酮试剂沸水浴10 min,检测波长λ=620 nm,回归方程A=-0.044 4+0.001 3X,r=0.999 54。结果补阳还伍汤中多糖的提取率为7.75%。结论微波辅助萃取大大缩短了提取时间,提高了收率,多糖的测定方法简便易行,准确可靠。

Abstract:Objective To extract the polysaccharides and determine content in Buyang Huanwu Tang.Methods The polysaccharides of Buyang Huanwu Tang were extracted by microwave assisted extraction(MAE).The water volume was 7 times of materals and extraction time was 20 minute with microwave power of 325w.The concentration of polysaccharides was dertermined by anthracenone colorimetry.Anthracenone reagent was added in boiling water,bathing for 10min,detection wavelength was at 620 nm,the regression equation Y=-0.044 4+0.001 3X,r=0.999 5.Results The extraction rate of polysaccharides in Buyang Huanwu Tang was 7.75%.Conclusion MAE shortens the extract time greatly and enhances the yield obviously.The method was convenient and feasible for determination of polysaccharides in the preparation.
