Abstract:OBJECTIVE: To investigate the gastrointestinal absorption of LMWH solution and emulsion in anesthetized rats. METHODE: LMWHs formulated as solution to emulsion were given by gastric garage or duodenal injection in anesthetized rats. Blood samples were collected and analyzed for anti-Xa, while anti-thrombosis activities were vestigated. RESULTS:At a dosage of 5000 Anti-Xa IU/kg,there was no significant change in plasma anti-Xa activity between gastric garage administration of LMWH solution and blank control; however, plasma anti-Xa activity was significantly increased after gastric garage administration of LMWH emulsion and significant anti-thrombosis activity was observed. In contrast, at the same dosage by duodenal injection, significant absorption was observed in both formulations, reaching peaks around 1 hour after administration. Significant anti-thrombosis activity was also observed. CONCLUSION: LMWH may be destroyed by gastric juice. But can be absorbed in duodenum at certain level. Formulation with monoolein or taurocholate can promote its absorption.
