压力负荷模型大鼠心肌解偶联蛋白-2基因表达与心肌纤维化的研究

摘    要:目的:探讨心肌线粒体解偶联蛋白-2(UCR2)在心肌纤维化发生、发展中的作用。方法:实验动物模型采用腹主动脉结扎法,对心肌UCP2mRNA检测采用RT—PCR方法,计算机凝胶成像分析其相对含量。心肌常规病理切片,sirius red染色,图像分析胶原含量。结果:(1)手术组大鼠的心重指数(CI)、腹主动脉平均压(MAP)持续升高;与假手术组比较,手术组CI术后4天开始差异有统计学意义(P〈0.05);而MAP术后1天起,两组差异有统计学意义(P〈0.05);(2)术后1天,手术组UCP,mRNA就升高,至实验结束始终处于较高水平状态,平均达假手术组的1.5倍左右;(3)手术组和假手术组大鼠比较,心肌血管周围胶原和管腔的面积比值(PVCA)在手术后1天即有显著升高(P〈0.05),以后逐步攀升;而心肌胶原容积分数(CVF)在手术后21天才有迅速增加(P〈0.05)。结论:UCP2参与了心肌纤维化的发生和发展,不仅是一个双向因素,而且反映心肌的功能储备。

Abstract:Objective:To investigate the role of uncoupling protein 2(UCP_2) in the myocardium fibrosis with pressure load. Methods:In this experiment, the abdomial aorta ligation was used to build the animal model, and the RT-PCR method was used to check the relative content of UCP_2 mRNA. The sections of myocardium were dyed by sirius red and compared by histopathological and computer image analyzer techniques. Results:(1)The cardiac index(CI) and the abdominal aorta pressure increased continuously; Compared with sham-operated group, the CI of the operated group significantly increased on the 4th day after operation(P<0.05); But the MAP of the two groups have difference on the 1st day after operation(P<0.05);(2)One day after operation, the expression of UCP_2 mRNA of the operated group increased. And it did not keep high level until the end of the experiment. On average, it increased 1.5-fold as much as the sham-operated group; (3)Compared with the sham-operated group, the PVCA of operated group increased evidently one day after operation(P<0.05), but the CVF of the operated group increased evidently on the 21st day after operation(P<0.05). Conclusion:The UCP_2 participates in the occurrence and the development of myocardium fibrosis, and it is not only a bidirection agent, but also a section of backlog for cardiac function.
