减肥胶囊减肥机理的分子水平研究

摘    要:目的从分子水平探讨减肥胶囊对营养性肥胖大鼠的减肥机理。方法采用组织原位杂交技术观察药物治疗组和模型对照组大鼠脂肪组织瘦素基因(Leptin mRNA),下丘脑神经肽Y基因(NPY mRNA)及解偶连蛋白2基因(UCP2 mRNA)表达水平,采用放射性免疫技术检测药物治疗组及非药物治疗组大鼠血清瘦素(Leptin)及神经肽Y(NPY)的含量。结果减肥剂量高、低剂量组均可降低大鼠血清Leptin水平,但与模型对照组比较,差异无显著性(P>0.05);用药后高、低剂量组大鼠血浆NPY水平均有降低,与模型对照组比较差异有显著性意义(P<0.05或P<0.01)。减肥胶囊高剂量组脂肪组织Leptin mRNA及下丘脑NPY mRNA表达水平下降,同模型对照组相比,差异有显著性意义(P<0.05或P<0.01);减肥胶囊高、低剂量组UCP2 mRNA水平均升高,与模型对照组比较,差异有显著性意义(P<0.05或P<0.01)。结论减肥胶囊的作用机理可能是影响了大鼠脂肪组织Leptin mRNA、下丘脑NPY及UCP2 mRNA基因的表达。

Abstract:Objective To study the mechanisms of Jianfei Capsule on alimentary obesity rats at molecular level.Methods The level of Leptin mRNA in adipose tissue and neusopeptide Y (NPY) mRNA and UCP2 mRNA in hypothalamus of rats in medicine treatment and model control groups were observed by in situ hybridization. The content of Leptin and NPY in medicine and non-medicine treatment groups were detected by radio immunoassay.Results As compared with model control group, the level of Leptin in high and low dosage of Jianfei Capsule groups decreased, but there was no significant difference (P>0.05); the level of NPY decreased but UCP2 mRNA increased significantly (P<0.01 or P<0.05); the level of Leptin mRNA in adipose tissue and NPY mRNA in hypothalamus in high dosage of Jianfei Capsule group decreased significantly (P<0.01 or P<0.05).Conclusion Jianfei Capsule can reduce weight of rats with alimentary obesity, which might be relevant to its regulating the expression of Leptin mRNA in adipose tissue and NPY mRNA and UCP2 mRNA in hypothalamus.
