大鼠子痫前期模型实验研究

目的:目前子痫前期是产科的主要疾病之一和孕产妇主要死亡原因之一。为研究子痫前期的病因、发病机制、治疗及预防方法,需建立子痫前期的实验动物模型。方法:用5 8只雌性Wistar大鼠采用机械缩窄子宫动脉的方法,经过9 0天观察,对大鼠血压值、24小时尿蛋白、血液生化、肾脏组织病理学改变等相关指标的观察分析。结果:大鼠子痫前期模型组(D组)血压描记曲线明显比B、C、A组增高而且脉压差大。D组平均动脉压(MAP)与B、C、A组比较均增高有非常显著差异(P<0.01)。B、C、A组比较无显著差异(P>0.05)。D组孕鼠胎仔及胎盘重量较B、C、A组明显减轻(P<0.01),死胎率显著增加(P<0.01)。D组24小时尿蛋白、CRE、BUN、NA+浓度与B、C、A组比较明显增高,有显著性差异(P<0.05)。本实验所显示孕鼠的高血压、24小时蛋白尿、IUGR、肾脏组织病理学改变,说明大鼠子痫前期模型与人类的子痫前期相似性。结论:成功的建立了Wistar大鼠子痫前期模型,为子痫前期病因学和临床治疗的研究提供了实验动物模型。研究初探为本实验研究积累了丰富资料,尤为在今后实验中应注意的细节问题,实验中大鼠子宫动脉狭窄范围与子痫前期疾病严重程度关系有待进一步探讨。

Experimental Study in Rat Model of Pre-eclampsia

Objective:Currently,pre-eclampsia is one of the major diseases in obstetrics and maternal major causes of death.To study the cause of pre-eclampsia,pathogenesis,treatment and prevention,there needed to establish an animal model of pre-eclampsia. Methods:With 58 female Wistar rats by mechanical narrowing of the uterine artery approach,after 90 days observation on blood pressure values,24h urine protein,blood,kidneys,histopathological changes and other related indicators were studied.Results:Rat model of preeclampsia group(D) blood pressure curve tracings was significantly higher than the other(B,C,A group),increased and Large pulse pressure.D group of mean arterial pressure(MAP) was significantly increased than the B,C,A group,there was a significant difference (P<0.01).B,C,A group showed no significant difference(P>0.05).D group of pregnant rats and fetal placental weight than the B,C,A significantly reduced(P<0.01),stillbirth rate was sigmificantly increased(P<0.01).D Group of 24-hour urine protein,CRE,BUN,NA + concentration were significantly higher than the B,C,A group,there was a significant difference(P<0.05).In this study,pregnant mice displayed high blood pressure,24-hour proteinuria,IUGR,renal histopathological changes,indicating that pre-eclampsia rat model of human pre-eclampsia similarity.Conclusion:Successful establishment of a Wistar rat model of pre-eclampsia,the etiology of pre-eclampsia and clinical studies provide an experimental animal model.Preliminary experimental study based research has accumulated a wealth of information,particularly in future experiments should pay attention to the details of the experiment in a narrow range of rat uterine artery disease and the severity of pre-eclampsia and the relationship to be further explored
