高甘油三酯血症对去卵巢大鼠股骨Fas/FasL表达的影响

摘    要:目的:探讨高甘油三酯(TG)血症对去卵巢大鼠股骨成骨细胞(OB)和破骨细胞(OC)中Fas、FasL的表达的影响。方法:将3月龄雌性SD大鼠40只,用果糖饲养复制高TG模型大鼠分为四组:(1)巢+果糖组;(2)去卵巢+果糖+非诺贝特(fenofibrate,FF)组;(3)去卵巢+普食组;④假手术+果糖组。12周后取股骨检测成骨细胞(OB)和破骨细胞(OC)中Fas、FasL的表达水平。结果:(1)去卵巢+果糖组的TG水平高于去卵巢+普食组(P<0.01),也高于去卵巢+果糖+FF组(P<0.01);(2)去卵巢+果糖组成骨细胞Fas、FasL的表达水平均高于去卵巢+果糖+FF组、去卵巢+普食组和假手术+果糖组(P<0.05);破骨细胞Fas、FasL的表达均低于其余三组(P<0.05)。结论:高TG血症使股骨组织中成骨细胞Fas、FasL表达增高,破骨细胞中Fas、FasL表达降低。

Effects of Hyper-triglyceride and Antilipemic Agents on Expressin of Fas/FasL in Femora of Ovariectomized Rats

Abstract:Objective:To investigate hypertriglyceridemia and antilipemic agents on the expression of Fas,FasL in ovariectomized rats.Methods:Forty female SD rats were randomly divided into 4 groups.(1)Ovariectomy + fructose(OVX+FO),(2) Ovariectomy + fructose +fenofibrate(OVX+FO+FF),(3)Ovariectomy + normal diet(OVX +ND),(4)Sham operation + fructose(Sham+FO).Twelve weeks after operation,all rats were sacrificed and right femora were taken.Then Fas.FasL expression were tested and analysed by pathological and immunohistochemical technology.Results:Hypertriglyceridemia down-regulated the expression of Fas.FasL on the osteoclasts but up-regulated Fas.FasL expression in osteoblasts;Fenofibrate up-regulated the expression of Fas.FasL expression in osteoclasts but down-regulated Fas.FasL expression in osteoblasts.Conclusions:Fructose can induce hypertriglyceridemia in ovariectomized rats which lead to down-regulate of Fas.FasL expression in osteoclasts and up-regulate Fas.FasL expression in osteoblasts.Hypertriglyceridemia may accelerate above changes and fenofibrate can prevent the adverse effect.
