芎附散对偏头痛模型大鼠行为学、血浆CGRP和NO的影响

摘    要:目的 观察芎附散对硝酸甘油诱发的偏头痛模型大鼠行为学、血降钙素基因相关肽(CGRP)和一氧化氮(NO)的影响.方法 将72只健康雄性SD大鼠随机分为空白对照组、模型组、盐酸氟桂利嗪组和芎附散组,每组18只,连续灌胃8天后,除空白对照组外,其余各组大鼠予以颈部皮下注射硝酸甘油复制实验性偏头痛模型.观察各组大鼠行为学变化1h,分别于1、2、4h三个时间点采集腹主动脉血测定CGRP和NO水平.结果 空白对照组、盐酸氟桂利嗪组和芎附散组大鼠1h行为学表现与模型组比较,差异有统计学意义(P<0.05).空白对照组、盐酸氟桂利嗪组和芎附散组大鼠1、2h两个时间点血CGRP水平与模型组比较差异无统计学意义(P>0.05),但三组4h血CGRP水平显著低于模型组(P<0.05).三组大鼠各时间点动脉血NO水平均低于模型组(P<0.05).结论 芎附散可减轻由硝酸甘油诱发的偏头痛模型大鼠症状,降低其血管活性物质CGRP和NO水平.

Abstract:Objective To observe the influence of Xiongfu powder on the behavioral manifestations and blood calci-tonin-gene related peptide(CGRP)and nitric oxide(NO)levels in migraine rats. Methods Seventy-two SD ratswere randomly divided into four groups blank control group, model group, flunarizine hydrochloride group, andXiongfu powder group. By intragastric administration for 8 consecutive days, the rats were injected subcutaneouslywith nitroglycerin to trigger migraine except for blank control group on day 8 and then abdominal aorta blood fromthe rats was drawn at 1,2, and 4 h after drug intervention to detect CGRP and NO levels. One-hour behavioralmanifestations were observed after drug intervention. Results One-hour behavioral manifestations were significantlydifferent in blank control, flunarizine hydrochloride, and Xiongfu power groups compared with the model group(P〈0.05). The CGRP level in blank control, flunarizine hydrochloride, and Xiongfu powder groups was markedly lowerthan that of model group at 4 h after drug intervention(P〈0.05), but no statistical difference was found betweenthe three groups and model group at 1 and 2 h(P〉0.05). The No levels in the 3 groups were much lower thanthat in model group at each time point(P〈0.05). Conclusion Xiongfu powder may relieve symptoms of rats follow-ing nitroglycerin infusion, meanwhile may reduce the CGRP and NO levels.
