靶向抑制CXCR7基因表达对结肠癌生长的影响

摘    要:目的探讨靶向抑制CXCR7基因表达对人结肠癌荷瘤裸鼠瘤体生长的影响。方法建立人结肠癌荷瘤鼠模型。将人结肠癌荷瘤裸鼠随机分为3组,治疗组:瘤体内注射LV-CXCR7shRNA 50μl;阴性对照组:瘤体内注射LV-shRNA negative control 50μl;空白对照组:瘤体内注射PBS 50μl。24天后测量瘤体的体积;处死动物,剥离瘤体,测量其重量。采用Westrn Blotting方法测定瘤体组织中CXCR7蛋白表达水平。结果治疗组瘤体体积显著缩小(P〈0.05)、重量显著减轻(P〈0.05)。治疗组瘤体组织中CXCR7蛋白表达水平显著降低(P〈0.05)。结论靶向抑制CXCR7基因的表达能显著抑制人结肠癌荷瘤裸鼠的瘤体生长,CXCR7是人结肠癌治疗潜在的分子靶标。

Abstract:Objective To explore the Growth effect on tumors of human colon tumor-bearing nude mice by inhibition expression of targetting-CXCR7 gene.Methods To establish the animal model of human colon tumor-bearing mice.Tumor-bearing nude mice were randomized into 3 groups and each group contained 8.Treatment groups:injecting 50 μL of LV-CXCR7shRNA into tumor;Negative control group:injecting 50 μl of LV-shRNA negative control into tumor;Blank control group:injecting 50 μl PBS into tumor.Tumor volumes were measured after 24 days.To sacrifice animals,remove tumors,measure tumor weights.CXCR7 protein of tumors were measured by Westrn Blotting.Results Tumor volume and weight of treatment group decreased significantly(P 0.05),respectively.The expression of CXCR7 protein in tumors of treatment group were down-regulated significantly(P0.05).Conclusion Inhibition expression of targetting-CXCR7 gene can effectively restrain the growth of tumor of human colon tumor-bearing nude mouse.CXCR7 is a potential molecular target for colon cancer therapy.
