胃癌及癌前病变中MGMT的表达及其意义

摘    要:目的:探讨DNA损伤修复基因产物-MGMT在胃癌及癌前病变中的表达及其意义。方法:采用免疫组化SP法检测MGMT在62例胃癌和42例胃黏膜异型增生组织中的表达。结果:MGMT蛋白在正常胃黏膜、轻度异型增生胃黏膜、中重度异型增生胃黏膜、早期胃癌、进展期胃癌中的阳性表达率分别为93.8%、94.4%、91.7%、66.7%、68.2%。MGMT蛋白在轻度异型增生胃黏膜的表达率明显高于早期胃癌及进展期胃癌(P<0.05);在中重度异型增生组中的表达率高于早期胃癌(P<0.05)。胃癌中MGMT表达与分化程度、淋巴结转移相关(P<0.05)。结论:MGMT基因可能在胃癌发生中起重要的作用,可能成为监测胃黏膜早期癌变及判断胃癌生物学行为的有用指标。

Abstract:Objective:To investigate the expression and significance of MGMT in gastric carcinoma and precancerous lesions.Methods:Immunohistochemical SP method was used to detect the expression of MGMT in 62 cases of gastric cancer and 42 cases of gastric dysplasia.Results:MGMT in normal mucosa、slight dysplasia、moderate and severe dysplasia、early gastric cancer,advanced gastric cancer,the positive expression rates were 93.8%,94.4%,91.7%,66.7%,68.2%,respectively.The positive rate of MGMT in early and advanced gastric carcinoma was lower than that in slight dysplasia(P<0.05).The positive rate of MGMT in moderate and severe gastric carcinoma was significantly higher than that in early gastric carcinoma(P<0.05).The expression of MGMT in gastric carcinoma tissue was closely corrected with the differentiation,lymph node metastasis(P<0.05).Conclusion:MGMT might play an important role in carcinogenesis of gastric mucosa and MGMT protein could be used as a marker to predict the carcinogenesis of gastric carcinoma and to judge the biological behavior of gastric carcinoma.
