氟伐他汀联合百令胶囊防治肾小球硬化实验研究

目的探讨氟伐他汀联合百令胶囊对防治肾小球硬化的作用。方法40只雄性SD大鼠分为4组,A组(n=9)为正常对照组,B组(n=10)、C组(n=10)、D组(n=11)采用阿霉素注射制备肾病综合征动物模型。A组(正常对照组):每日灌服自来水3.0ml;B组(肾病对照组):每日灌服自来水3.0ml;C组(百令胶囊组):每日灌服百令胶囊粉2.0mg·kg-1·d-1;D组(联合用药组):每日灌服百令胶囊粉2.0mg·kg-1·d-1,氟伐他汀2.0mg·kg-1·d-1。各组均灌服6周后,观察血清脂谱、血清白蛋白(Alb)、肌酐(Scr)、尿素氮(BUN)、24小时尿蛋白定量(24hPro)及肾脏病理改变。结果D组血脂谱明显低于B、C组(P<0.01),Alb明显高于B组(P<0.01),而与C组无显著差异,同时C、D2组肾组织损伤也明显减轻,免疫组化和计算机定量分析进一步显示细胞外基质(III、IV型胶原,纤维连接蛋白,层黏蛋白)肾小球内聚积明显少于B组。结论氟伐他汀和百令胶囊联合用药能更有效的降低血脂,减少白蛋白排泄,减缓肾小球硬化,防治肾纤维化,保护肾功能。

Experimental study of prevention and treatment of glomerulosclerosis by fluvastatin combined with Bailing capsule

Objective To investigate the prevention and treatment of fluvastatin combined with Bailing capsule on glomerulosclerosis.Methods The rats were divided into four groups, group A (normal control group) administered with water through gastric tube 3.0 ml -1·d -1 for 6 weeks,group B was nephropathy control group,group C (Bailing capsule group) using Bailing capsule 2.0 mg·kg -1·d -1, group D (combined treatment group) using fluvastatin 2.0 mg·kg -1·d -1 and Bailing capsule 2.0 mg·kg -1·d -1 for 6 weeks.The blood lipids,Alb,Scr,BUN,24h Pro and the pathological changes of kidney were observed.Results The choloesterol,triglycerides,very low lipoprotein,high density lipoprotein and 24 h Pro in combined treatment group were significantly lower than those of nephropathy control group (P<0.01),however the serum albumin was higher than that of nephropathy group (P<0.01),moreover,the renal damage was also less significant in combined treatment group.Light microscope and immunohistochemical examination revealed that less accumulation of extracellular matrix including collagen III,IV,laminine and fibronectin in combined treatment group, as compare with that in nephropathy control group.Conclusion Fluvastatin combined with Bailing capsule may play an important role in the pathophysiological progression of renal disease, which may retard the progression of glomerulosclerosis and thus protect renal function.
