高压氧对去卵巢大鼠泪液分泌及泪腺形态学的影响

摘    要:目的 建立去卵巢大鼠干眼症模型,并探讨高压氧对其泪液分泌及泪腺形态学的影响.方法 32只6个月龄雌性Wistar大鼠按随机数字表法分成4组(每组8只):假手术组、去势组、雌激素组与高压氧组.造模成功1周后给予治疗干预,12周后行Schimer Ⅰ试验、泪膜破裂时间检测、角膜荧光素染色检测及泪腺形态学观察.结果 去势组泪液分泌量与泪膜破裂时间均明显低于假手术组,且大鼠角膜干燥无光泽,上皮点状甚至片状缺失,荧光素染色阳性,泪腺形态学显示腺泡泡状黏液消失,分泌低下,表明造模成功;雌激素组[(6.14±2.13)ml,(11.21±1.99)s]与高压氧组[(5.16±1.79)ml,(10.86±1.78)s]泪液分泌量与泪膜破裂时间均明显高于去势组[(3.25±1.66)ml,(8.97±2.01)s](P<0.05),且大鼠角膜透明,偶见点状荧光素染色,泪腺形态学显示分泌旺盛.结论 高压氧可减轻泪腺炎症反应,促进泪液分泌,在一定程度上可以减轻干眼症模型的泪液分泌过少症状.

Abstract:Objective To develop a xerophthalmia model and observe the effects of hyperbaric oxygen (HBO) on tear secretion and morphology of lacrimal gland in ovarientomized rats. Methods Thirty-two female Wistar rats were randomized into 4 groups, each consisting of 8 animals, i.e. the model group, the control group, the EB group and the HBO group. Therapeutic intervention was made 1 week after the development of the model. Schirmer' s tests, tear film break - up time, corneal fluorescence staining were performed and morphology of lacrimal gland was observed after 12 weeks. Results The amount of tear secretion and tear film break- up time were significantly lower or shorter in the model group than those in the control group, with cornea being dry and dim. And the corneal fluorescence staining was positive in the model group. Morphologic observation on the lacrimal glands indicated that bubble - like mucus was absent, with little secretion of tears,indicating that the animal model was successfully developed. The anount of tear secretion and tear film breakup time in the EB group and the HBO group, were significantly higher or longer than those in the model group.Furthermore, fluorescence staining showed that the corneal was fairly transparent and morphologic observation indicated that secretion of lacrimal glands was normal. Conclusions HBO could alleviate the inflammatory reaction of lacrimal glands, enhance the secretion of tears and could, to a certain extent, relieve symptoms of oligodacrya in xerophthalmia.
