不同镀膜磁体对牙龈成纤维细胞活性及mTOR表达的影响

摘    要:目的评价各种镀膜钕铁硼磁体的生物相容性。方法将托槽、不同镀膜及未镀膜磁体加入DMEM中制备成浸提液,分别以5%和10%浓度加入培养的大鼠牙龈成纤维细胞,以AlamarBlueTM法测细胞活性。对加入10%浸提液的培养细胞做雷帕霉素靶蛋白(mTOR)的免疫印记分析。结果结果显示5%托槽、镀氮化钛磁体组的AlamarBlue还原率分别为63.0±3.4%、66.5±3.0%,与对照组64.2±3.4%无差异(P>0.05);10%托槽、镀氮化钛磁体组的还原率分别为62.0±2.17%、64.9±0.5%,与对照组比较65.7±1.0%无差异(P>0.05);而其余各组培养细胞的AlamarBlue还原率与对照组有显著性差异(P<0.05)。免疫印记结果显示,mTOR表达的平均灰度值托槽组77.0±2.85与镀氮化钛组81.2±2.60无显著性差异(P>0.05);镀镍组40.5±1.94,镀铜组15.0±2.01和未镀膜磁体组24.7±2.10与托槽组差异显著(P<0.01)。结论镀氮化钛钕铁硼磁体具有良好的生物相容性。

Abstract:Objective To investigate the biocompatibility of different coating NdFeB magnets. Methods Brackets ,different coating magnets and bare magnets were incubated in DMEM to make solutions, 5%and 10% those solutions were added into cultured rat gingival fibroblast cells. AlamarBlue  TM was used to examine the changes of cell activity . Western blotting was used to examine the expression of mammalian target of rapamycin( mTOR) in cells cultured with 10% solutions .Results The percentage of reduced AlamarBlue in 5% brackets, titanium-nitride magnets groups were 63.0±3.4%,66.5±3.0%,without obvious difference compared with control group 64.2±3.4% (P> 0.05). The percentage of reduced AlamarBlue in 10% bracket, titanium-nitride magnet groups were 62.0±2.17%,64.9±0.5%,without obvious difference compared with control group 65.7±1.0% (P> 0.05). The percentage of reduced AlamarBlue in nickle, copper, bared magnet groups had significant difference compared with control groups(P< 0.05).Western blotting showed that there was no significant difference for average gray value of mTOR between bracket 77.0±2.85 and titanium-nitride 81.2±2.60 (P<0.05),but there was significant difference in nickle group 40.5±1.94,copper group 15.0±2.01, bared magnet group 24.7±2.10 compared with bracket group(P<0.01).Conclusion Magnet coated titanium nitride has good biocompatibility.
