RhD基因检测方法在Rh阴性血型基因鉴定中的应用

摘    要:目的:探讨RhD基因检测方法在中国汉族人Rh阴血型鉴定中的应用价值。方法:对于68例各种表型的Rh阴性中国汉族样本,用扩增RhD基因外显子3,4,5,7,10的5种方法,检测RhD基因的存在与否。结果:68例样本,总阴性率为52.95,其中29例含有C抗原的Rh阴样性本,5种方法鉴定皆为阴性的仅有1例(3.4%),而表型为C阴性的38例样本,34例(89.5%)为阴性。结论:以检测RhD基因存在与否为基础的Rh阴性血型基因鉴定方法,不适用于C抗原阳性的中国汉族Rh阴性血型基因鉴定,仅有扩增RhD外子3,4,10的方法,适用于含C抗原的Rh阴性血型的基因鉴定。

Application of RhD gene detection method in Rh negative blood group gene identification
Abstract:Objective To evaluate the application value of RhD gen e detection method in Rh negative blood group in Chinese Han population. Method Five methods were used by which RhD gene exon 3,4,5,7 an d 10 were amplified respectively to detect the RhD gene in 68 Chinese Han Rh neg ative phenotype samples. Results The RhD gene negative rate detected by all of five meth ods was 52. 9% in 68 samples.In 29 with C antigen positive samples, only 1 sampl e (3. 4%) was negative, but in another 38 with C antigen negative samples, 34 sa mples (89. 5%) were negative.Conclusion The Rh negative blood g roup genotyping methods based on detecting Rh D gene are not suitable for Rh neg ative phenotype of C antigen positive in Chinese Han population.Only the methods which amplify RhD gene exon 3,4,10 respectively might suitable for C antigen ne gative samples.
