摘    要:目的:观察软脉冲剂对mox-LDL诱导血管平滑肌细胞(VSMC)增殖的影响。方法:培养的血管平滑肌细胞分为对照组、mox-LDL组和用药组(mox-LDL+不同剂量软脉冲剂),应用MTT法和流式细胞仪分别测定细胞含量和细胞周期,用硫代巴比妥酸比色法测定细胞培养液中丙二醛(MDA)的水平。结果:软脉冲剂能明显降低细胞培养液中MDA的水平,MTT检测显示软脉冲剂具有浓度依赖性抑制VSMC的增殖的的作用,流式细胞仪检测表明,用药组(S+G2M)期降低。结论:软脉冲剂能抑制mox-LDL刺激的SMC的增殖,并具有抗脂质过氧化作用。

Abstract:Objective:To investigate the effect of Ruanmai Granulum on proliferation and lipid peroxidation oxidized of bovine vascular smooth muscle cells(VSMC).Method:The subjects were divided into three groups:control group,mildly Low Density Lipoprotein(mox-LDL)group and Ruanmai Granulum Treatment group who were treated with three different dosages.Using experimental models of proliferation of cultured bovine VSMC induced by mildly oxidized-low density lipoprotein,the level of MDA was determined with thiobarbituric acid reactive substance and the number of bovine VSMC was measured with MTT and the cell cycle phase was measured by a flow cytometer.Result:Ruanmai Granulum markedly reduced the level of MDA in the medium and inhibited MTT metabolism of VSMC.Ruanmai Granulum was shown to suppress VSMC proliferation in a concentration-dependent fashion.Ruanmai Granulum reduced the(S+G2M)block in cell cycle traverse.Conclusion:Ruanmai Granulum can inhibit the proliferation of VSMC.The inhibitory effect may be related to the inhibition of lipid peroxidation.
