大孔吸附树脂富集纯化川贝母总生物碱的工艺研究

摘    要:目的:研究大孔吸附树脂富集纯化川贝母总生物碱的工艺参数及条件。方法:7种不同类型的大孔吸附树脂被研究,以总生物碱的吸附率及解吸率为考察指标,探索吸附和解吸的条件,筛选出纯化总生物碱的最佳工艺。结果:HPD-100大孔树脂对川贝母总生物碱有较好的富集纯化性能。最佳工艺条件为:上样药液质量浓度为0.293g生药/mL,pH值为11.6,6BV的80%乙醇洗脱总生物碱,洗脱体积流量为2BV/h。按照此工艺条件纯化的总生物碱纯度达到51.92%,收率76.65%。结论:建立HPD-100大孔吸附树脂富集纯化川贝母总生物碱的方法简单,可靠,重现性好,适合富集纯化川贝母总生物碱,为其它种贝母总生物碱的富集纯化提供参考。

Abstract:Objective To study technological parameters of enriching and purifying total alkaloids from Fritillariae Cirrhosae Bulbus by macroporous resins.Methods The adsorptive and desorption ratio of the total alkaloids was determined by UV spectra.The optimized macroporous resins were selected by studying 7 kinds of macroporous resins.The optimized technological parameter was screened out by further studying absorption and desorption.Results HPD-100 macroporous resins was selected as the best material in enrichment and purification of total alkaloids for Fritillariae Cirrhosae Bulbus.The optimized technological paramaters was 0.293g/mL concentration,pH value 11.6,delution by 6BV,80% alcohol in 2BV/h.The content of total alkaloids obtained was 51.92%,and the yield was 76.65%.Conclusion Using HPD-100 macroporous resins to enrich total alkaloids in Fritillariae Cirrhosa Bulbus is easy,relable and good reproductively,which could also provide a reference for other Fritillariae species.
