Abstract:Aim To investigate the effects of growth inhibitio n of different concentrations of mitomycin polybutylcyanoacrylate magnetic nanpart icles(MMC-PBCA-MNPS), PBCA-MNPS and MF (magnetic fluids) and MMC on normal hu man liver cell strain (L-02 cells) in vitro.Methods MMC-P BCA-MNPS and PBCA-MNPS were prepared by emulsion polymerization method, respectively. The influences of growth inhibition of different concentrations of MMC-PBCA-MNPS,PBCA-MNPS, MF and MMC on L-02 cells were studied by MTT colorimetric assay; the activity of lactate dehydrogenase (LDH) was detected by automatic biochemy-instrument. Results The relative inhibition rates (RIR%) of different concentrations of MMC-PBCA-MNPS for L-02 cells were:10.0%,15.1%,25.1%,32.1%,39.9% and 45.0%;PBCA-MNPS: 1.8%,5.2%,10.0%,12.9%,15.1% and 29.9%;MMC: 22.1%,35.1%,46.1%,62.0%,80.1% and 88.9%.MF did not significantly inhibit the growth of L-02 cells, meanwhile they were in favor of the growth of L-02 cells.Judging by relative growth rate (RGR%), the cytotoxicities grade of MMC-PBCA-MNPS, PBCA-MNPS, MF and MMC for L-02 cells were 1, 1, 0 and 4 grade, respectively. The activities of LDH were as follow: MMC-PBCA-MNPS: 117.00±16.00~354.0±14.18; PBCA-MNPS: 106.00±13.00~186.00±15.72; MF: 35.00±12.49~34.00±6.56 and MMC: 377.00±10.82~471.00±9.85; and blank: 34.00±5.29, P<0.05;P<0.01.Conclusion The relative degree of growth inhibition for L-02 cell strain is as follows: MMC>MMC-PBCA-MNPS>PBCA-MNPS>MF.
