乙型和丙型肝炎病毒双重感染肝组织病毒标志物的检测

为观察乙型肝炎和丙型肝炎双重感染的尸体肝组织中两种病毒核酸及抗原的分布，对１５例ＨＢＶ与ＨＣＶ双重感染的尸检肝组织用免疫组织化学法检测ＨＢｓＡｇ、ＨＢｃＡｇ和ＨＣＡｇ，Ｄｉｇｏｘｉｇｅｎｉｎ标记的探针原位杂交法分别检测ＨＢＶ ＤＮＡ和ＨＣＶ ＲＮＡ。结果：ＨＢｓＡｇ、ＨＢｃＡｇ、ＨＢＶ ＤＮＡ、ＨＣＡｇ和ＨＣＶ ＲＮＡ的阳性数分别为１２／１５（８０．０％）、１０／１５（６６．７％）、９／１５（６０

Detection of Intrahepatic Viral Markers in Patients with HBV and HCV Dual Infection

In 15 autopsy liver tissues with HBV and HCV dual infection, HBV DNA, HBsAg, HBcAg, HCAg and HCV RNA were detected respectively. Of the 15 cases,11 had liver cirrhosis (LC). 3 had chronic severe hepatitis (CSH) and 1 with subacute severe hepatitis (SSH). Viral antigens were stained by immunohistochemistry. Using digoxigenin,HBV DNA probe was labelled by random priming and HCV cDNA probe was labelled by polymerase chain reaction assay. Then, HBV DNA and HCV RNA were detected by in situ hybridization. Both HCAg and HCV RNA were observed in cytoplasm or in nuclei of hepatocytes. Most patients with LC in this study had long history of HBV infection and transfusion or blood products infusion for many times. The positive cases of 11 LC were : 8 had HBsAg;6 had HBcAg;5 had HBV DNA;8 had HCAg and 9 had HCV RNA. It seemed that the cases with HCV RNA and its antigen were a little more than those with HBcAg and HBV DNA. Maybe it correlated to time order .of the dual infection occurrence. These five markers were all present in 3 CSH samples. One case with SSH had HBV DNA and its two antigens,but no detectable HCV in the liver although serum anti-HCV was positive.
