癌基因STAT3与结直肠癌恶性程度关系的研究

目的:通过研究癌基因STAT3在结直肠癌及癌旁组织中的表达,探讨其在结直肠癌发病机制中的作用。方法:采用免疫组化MaxVisionTM法检测40例结直肠癌组织、37例癌旁组织和30例正常肠粘膜组织中STAT3蛋白的表达,并对结直肠癌组织中STAT3蛋白的表达情况与不同临床病理特征进行统计学分析。结果:在结直肠癌和癌旁组织中,STAT3蛋白的表达差异无显著性(P>0.05)。相对于正常肠粘膜组织,结直肠癌和癌旁组织的STAT3蛋白表达有显著差异(P<0.05)。STAT3蛋白的表达水平与结直肠癌的分化程度、临床分期及淋巴结转移有关(P<0.05).结论:STAT3高表达可能是结直肠癌发病机制中的早期事件。在癌旁组织中,那些STAT3蛋白阳性的组织细胞可能是潜在的癌前细胞。检测结直肠癌中STAT3蛋白的表达可以反映肿瘤的恶性程度。

The research on the relationship of oncogene STAT3 and malignancy degree in colorectal carcinoma

Objective:By researching the expression oncogene STAT3 in colorectal carcinoma(CRC)tissues and para- carcinoma tissues,to explore its role mechanism in tumorigenesis of CRC.Methods:The protein expressions of STAT3 were detected in 40 CRC tissues,37 para-carcinoma tissues and 30 normal colorectal mucosa tissues by Max VisionTM immunohistochemistry,and the relationship of the expression of STAT3 in CRC with various clinico- patholoical characteristics was analyzed statistically.Results:There was no significant difference between the expres- sion of STAT3 protein in CRC tissues and para-carcinoma tissues(P>0.05).Compared with normal colorectal mu- cosa tissues,the expression of STAT3 protein in CRC tissues and para-carcinoma were significantly higher(P<0.05).Overexpression of STAT3 protein was correlated with differentiation,clinical stage and lymph node metasta- sis in CRC(P<0.05).Conclusions:Overexpression of STAT3 may be an early accidence in the pathogenesis of CRC.In para-carcinoma tissues,those STAT3-positive may be potential pre-cancerous cells.Determination of STAT3 protein can be used to indicate the malignancy degree of CRC.
