阻断白三烯B4/白三烯B4受体1途径在川崎病治疗中的作用

目的观察白三烯B4（LTB4,leukotriene B4）在川崎病患者血清中的表达及阻断白三烯B4/白三烯B4受体1（LTB4-BLT1）途径对内皮细胞游走的影响,以探讨阻断LTB4-BLT1途径在川崎病治疗中的意义。方法用酶联免疫吸附试验（Enzyme-linked immunosorbent assay,ELISA）检测川崎病急性期及丙种球蛋白治疗后患儿的LTB4血清含量;用改良Boyden法检测加入BLT1拮抗剂U-75302后的血清对内皮细胞游走的影响。结果在川崎病急性期,血清LTB4含量明显增高,用丙种球蛋白治疗后下降;在加入BLT1拮抗剂后使得上述血清对内皮细胞的游走作用明显降低。结论 LTB4-BLT1途径参与川崎病血管炎性损伤过程,阻断LTB4-BLT1途径可能成为治疗川崎病血管内皮损伤的新靶点。

Blocking the LTB4-BLT1 pathway in the curing Kawasaki disease

Objective To investigate the effect of leukotriene B4 in the patients with Kawasaki disease.Meanwhile,the effects of human umbilical vein cell migration by blocking the LTB4-BLT1 pathway were observed.Methods The concentration of LTB4 in the serum from patients with Kawasaki disease was observed in a cure stage before and after IVIG.The migration of endothelial cells in plasma with U-75302 was determined by modified Boyden chamber methods.Results The level of LTB4 in Kawasaki disease was significantly higher than that in normal controls.Migration index showed decrease after blocking the LTB4-BLT1 pathway.Conclusion It is thought that LTB4-BLT1 pathway is involved in vascular damage in Kawasaki disease,blocking the LTB4-BLT1 pathway may be a new way to cure Kawasaki disease.
