多巴胺对PC12细胞凋亡的诱导作用

目的 研究外源性多巴胺对多巴胺能神经元的毒性作用，方法 采用体外培养的PC12细胞为模型，以终浓度0.45mmol/L的多巴胺对细胞进行诱导。结果 在透射电镜下可见培养的PC12细胞核染色质明显固缩、断裂，原位末端标记技术显示胞核内散在分布断裂DNA片段，流式细胞仪检测到DNA周期中G0-G1期前出现明显的亚二倍体峰，琼脂糖凝胶电泳呈现典型的“梯状”带等细胞凋亡的特征性改变。结论 外源性DA可诱导PC12细胞凋亡。

Dopamine Induced Apoptosis in PC12 Cells

Objective To study the toxicity of exogenous dopamine(DA) in dopaminergic neurons. Methods The PC12 cell was used as a cell model. Apoptosis was induced by DA. Results {After} addition 0 45 mmol/L DA, the PC12 cell showed condensed nucleus and chromatin under transmission electric microscope or detected by TUNEL. Meanwhile, there was a marked subdiploid peak before G-0|G-1 on DNA cycle with flow cytometry, and a very clear DNA ladder could be detected by gel electrophoresis. Conclusion The exogenous dopamine can induce apoptosis in PC12 cells.;
