磷酸肌酸钠对急性心肌梗死大鼠心室肌细胞动作电位和L型钙电流的影响

摘    要:目的 探讨磷酸肌酸钠对急性心肌梗死大鼠心室肌细胞动作电位(AP)和L型钙电流(Ica-L)的影响.方法 采用结扎大鼠左前降支的方法制备急性心肌梗死大鼠模型,应用膜片钳技术中电流钳记录急性心肌梗死大鼠游离心室肌细胞动作电位,用全细胞膜片钳技术记录急性心肌梗死大鼠游离心室肌细胞离子流.结果 磷酸肌酸钠组急性心肌梗死大鼠的心室肌细胞与对照组心室肌细胞相比,动作电位幅值(APA)下降,差异有统计学意义(P<0.01,n=7),动作电位时程(APD)有明显延长(P<0.01,n=7);磷酸肌酸钠明显增加急性心肌梗死大鼠的心室肌细胞L-型钙电流幅值(P<0.01,n=7).结论 磷酸肌酸钠降低急性心肌梗死大鼠动作电位幅值(APA),延长动作电位时程(APD);磷酸肌酸钠增加急性心肌梗死大鼠的心室肌细胞L-型钙电流幅值.

Abstract:Objective To study the effect of Creatine phosphate sodium (CPS) on the action potential (AP) and the L-calcium channel currents (Ica-L) of ventricular myocytes after the acute myocardial infarction Methods AP tracing were recorded by whole cell membrane current clamp and ICa-L were recorded by whole cell membrane vohage clamp after the rats were subjected to left anterior descending coronary artery ligation. Results The Creatine phosphate sodium (CPS) reduce the action potential Amplitude (APA)(P〈0.01, n=7) and prolong the action potential (P〈0.01, n=7) duration of the ventricular myocyte of the acute myocardial infarction (AMI) rats with the control significantly. Creatine phosphate sodium(CPS) increase the L-calcium channel currents(Ic~_L) of ventricular myocytes of AMI rats with the control significantly (P〈0.01, n=7). Conclusion The Creatine phosphate sodium (CPS)reduce the action potential Amplitude (APA)and prolong the action potential duration the ventricular myocyte of the AMI rats, the Creatine phosphate sodium(CPS)increase the L-calcium channel currents (ICa-L) of ventricular myocytes of AMI.
