针刺对大鼠缺血再灌注后脑细胞内钙稳态的影响及脑保护的机制探讨

摘    要:目的探讨针刺对抗脑缺血再灌注所致脑细胞凋亡的机制。方法采用线栓法制作SD大鼠脑缺血再灌注动物模型,于脑缺血30m in再灌注30m in时给予针刺干预,于再灌注72h处死动物取材,分别作脑细胞内游离钙、CGRP和脑细胞凋亡的检测。结果针刺干预组与单纯脑缺血再灌注组比较,脑细胞内游离钙明显降低(P<0.05),CGRP的表达明显增强(P<0.05),脑细胞凋亡数目明显减少(P<0.05)。结论针刺通过调节钙稳态及介导CGRP的分泌,减少脑缺血再灌注后脑细胞的凋亡,增强脑组织对脑缺血缺氧的耐受性。

Abstract:Objective To study the role of electroacupuncture in antagonizing neuronal apoptosis in rats with cerebral ischemia and reperfusion.Methods Model of cerebral ischemia was used by middle cerebral artery occlusion(MCAO).Electroacupuncture were given to rats when cerebral ischemia lasted for 30 minutes and then reperfused for 30minutes.Rats were randomly allocated to three groups:(1)sham operation(sham);(2)ischemic and reperfusion(I/R);(3)I/R(I/R EA) followed by electroacupuncture.The introcelluar liberated calcium,CGPR and neuronal apoptosis were determined.Results In electroacupunctured rats(I/R EA),the concentration of introcelluar liberated calcium and the expression of CGRP were significantly decreased(P<0.05),while the number of apoptosis neuronal cells was significanty decreased(P<0.05),compared with ischemia and reperfusion rats alone.Conclusion Electroacupuncture enhances the tolerance of cerebral tissue for hypoxia via regulating the homeostasis of calcium,promoting the secretion of CGRP and decreasing apoptosis cerebral cells resulted by cerebral ischemia reperfusion.
