针刺对脑缺血胶质增生的影响

摘    要:目的:通过针刺对脑缺血后星形胶质细胞增殖进行干预,以探讨其对胶质增生的影响。方法:建立大鼠大脑中动脉阻塞(MCAO)再灌注模型,应用免疫组化和免疫印迹方法检测2 d和7 d后损伤侧海马细胞周期蛋白依赖性蛋白激酶4(CDK4)和胶质纤维酸性蛋白(GFAP)的表达。观察针刺对胶质增生的影响。结果:MCAO后2d,损伤侧海马CDK4蛋白表达明显增强(P<0.01),MCAO后7 d,损伤侧海马GFAP蛋白的表达明显增强(P<0.01),给予针刺后,可抑制CDK4蛋白和GFAP蛋白表达(P<0.01)。结论:针刺可抑制胶质增生,从而有可能为神经再生和功能重建提供更有利的生存环境。其机制可能与其调控细胞周期有关。

Abstract:Objective: To investigate the effects of acupuncture on astrocyte proliferation in hippocampus after focal cerebral ischemia reperfusion in rats.Methods: Ischemia/reperfusion injury was induced by temporary middle cerebral artery occlusion(MCAO).Acupuncture was administrated once a day for 2 and 7 days after MCAO,the expression level of CDK4 and glial fibrillary acidic protein(GFAP) was analyzed by Immunohistochemistry.Results: Two days after cerebral ischemia reperfusion,the expression of CDK4 increased significantly(P<0.01).And 7 days later,that of GFAP increased significantly(P<0.01) too.Acupuncture could inhibit the expression of CDK4 and GFAP(P<0.01).Conclusion: By regulating cell cycle,acupuncture may inhibit the glial proliferation,which could contribute to the regeneration of nerve and recovery of function.
