痛风通络方对尿酸钠诱导急性痛风性关节炎的抗炎镇痛研究

目的:探讨痛风通络方对尿酸钠诱导大鼠急性痛风性关节炎的抗炎镇痛作用。方法:取60只大鼠,随机分成6组,对照组、模型组、吲哚美辛组、痛风通络方高、中、低剂量组,每组10只。除空白组外,其余5组采用尿酸钠晶体溶液踝关节注射建立急性痛风性关节炎模型。以痛风通络方与吲哚美辛对比治疗,观察大鼠步态,测定关节肿胀度,检测大鼠外周血白细胞数量变化,ELISA方法检测血清IL-1、IL-6、TNF水平。结果:痛风通络方高、中、低剂量在致炎后各时间点均能不同程度的抑制尿酸钠所致大鼠踝关节肿胀程度,降低白细胞含量,降低炎性因子IL-1、IL-6、TNF含量,与模型对照组比较差异有显著性(P<0.05)。结论:通用痛风方对尿酸钠所致急性痛风性关节炎症有较好的抗炎镇痛作用,其作用机制可能与降低炎症因子IL-1、IL-6、TNF水平有关。

Experimental Study of Gout Tongluo Decoction in Treatment of Acute Gouty Arthritis

Objective ： To study the anti - inflammatory and analgesic effects of Gout Tongluo Decoction in the treatment of uric acid sodium salt （GA） induced acute gouty arthritis in rats. Methods：60 rats were randomly divided into six groups, the control group,the model group, indomethacin group, Gout Tongluo Decoction high dose group, middle dose group, low dose group, 10 in each group. In addition to the control group, the remaining five groups were given ankle injection of sodium urate crystal solution to make the acute gouty arthritis model. Gout Tongluo Decoction compared with indomethacin treatment, rats were measured the de- gree of joint swelling, the quantity and concentration of rat peripheral white blood cells and neutrophils. ELISA was used to detect serum IL - 1, IL - 6 and TNF. Results ： In Gout Tongluo Decoction three groups, inflammatory each time points had varying degrees of inhibition of sodium urate inducing the ankle pain, reducing swelling degree of lower gait score and lowering inflammatory factors IL - 1, IL - 6 and TNF contents. Compared with the model group, the difference was significant （ P 〈 0. 05 ）. Conclusion ： Gout Tongluo Decoction has better therapeutic effect to sodium urate gouty arthritis disease and its mechanism may reduce the inflammatory cytokines IL - 1, IL - 6 and TNF levels.
