大功率氦氖激光治疗增生性瘢痕的实验研究

摘    要:目的建立增生性瘢痕动物模型,探讨大功率氦氖激光对增生性瘢痕的治疗作用及其机制.方法用圆形钻刀在兔耳腹面造成6mm直径的全层皮肤缺损创面32个,术后21d对其中10个突出瘢痕进行氦氖激光照射(输出功率120mW,光斑直径6mm,每次30min,隔日1次),另10个突出瘢痕作为对照,术后35d检测瘢痕组织中的细胞凋亡率(TUNEL技术)和PCNA蛋白表达(免疫组化).结果氦氖激光照射组瘢痕厚度变薄,明显小于对照组(P<0.05);TUNEL技术显示照射组瘢痕组织中细胞凋亡率增加,明显大于对照组(P<0.05),免疫组化表明照射组瘢痕组织中PCNA蛋白表达下调,显著低于对照组(P<0.05).结论大功率氦氖激光能抑制细胞增生性瘢痕的进程,原因可能与氦氖激光抑制细胞增殖、诱导细胞凋亡有关.

Abstract:Objective To explore the therapeutic effects and mechanisms of high-power He-Ne laser irradiation on hyperplastic scar (HS) in rabbits by setting up hyperplastic scar animal model.Methods 32 round wounds (diameter of 6 millimeter) with whole skin layer loss were made on ventral side of rabbits' ears using a trephine.21 days after the operation,10 projecting scars were irradiated with He-Ne laser (power of 120 mW,spot diameter of 6 millimeter) for 30 minutes every other day,altogether two weeks.Another 10 projecting scars were as control group.35 days later, cell apoptosis rate and PCNA protein expression of the scar histology were measured by TUNEL technique and immunohistochemistry technique respectively.Results In He-Ne laser group,scars became much thinner than those in control group(P<0.05).Cell apoptosis rate increased greatly as well as PCNA protein expression decreased a lot.There were significant difference between the two groups(both P<0.05). Conclusion It shows that high-power He-Ne laser could inhibit the growth process of HS in rabbits by inducing cell apoptosis and inhibiting cell proliferation probably.
