仔猪腹泻基因工程疫苗在环境中稳定性的初步研究

摘    要:观察不同辐射强度的自然光照射下,对不锈钢表面上的仔猪腹泻基因工程疫苗(MM3)和大肠杆菌野生株(8099)存活的影响.结果表明,在日光总辐射条件下.暴在10min,累积辐射0.476MJ/m2.MM3同8099的存活率比值为113.9;散射条件下.暴露80min,累积散射0.897MJ/m2,MM3同8099的存活率比值为1:11.2;黑暗条件下,暴露80min,MM3同8099的存活率比值为1:8.0。t检验.无论日光总辐射、散射还是黑暗环境,差别都高度显著(P<0.01).用含有氯霉素的LB培养基测定日光总辐射对MM3质粒稳定性的影响,未发现有质粒丢失.

Abstract:In this study,we observed the effect of solar radiation on the survival of the genetically live bacterial vaccine of preventing diarrhae of piglets(MM3)and the wild strain 8099 of Escherichia Coli(8099)on the smooth stainless steel suface.The results indicate that when the direct exposure to solar radiation was extended to 10min(total radiation dose:0.476MJ/m2),the survival ratio between the MM3 and 8099 was 1:3.9.In the diffusive condtion, the ratio of the survival percentage between the MM3 and 8099 was 1:11.2 when exposed for 80min(total diffusion dose:0.897MJ/m2).In the dark condition,the ratio of the survival percentage between the MM3 and 8099 was1:8.0,for the exposure time of 80 min,in the above the condition,the ratios of the survival percentage were significantly different(P<0.01 paired test)and no loss of the plasmid of the MM3 was detected in the solar radiation condition by using LB medium that contained chlormycetin.
