新型维甲酸对乳腺癌细胞周期蛋白D1、E表达的影响

摘    要:目的 研究新型维甲酸AHPN对乳腺癌细胞的生长抑制的可能机制。方法 应用细胞生长抑制试验及Western印迹法,研究新型维甲酸对乳腺癌细胞株MDA-MB-435细胞的生长抑制现象、不同时间点细胞内周期蛋白D1、周期蛋白E蛋白表达情况。结果 AHPN可有效地抑制MDA-MB-435细胞的生长,周期蛋白D1的蛋白表达随药物的作用时间的延长而逐渐降低,但周期蛋白E的蛋白表达却无明显的改变。结论 AHPN可有效地抑制人乳腺癌细胞株MDA-MB-435细胞的生长,这种抑制作用可能通过降低周期蛋白D1的蛋白表达实现,而与周期蛋白E的蛋白表达量无明显关系。

Effects of a Novel Retinoid on Cyclin D1 and E in Breast Cancer Cells
Abstract:Purpose To investigate the mechanism of the growth inhibition induced by a novel retinoid AHPN in breast cancer cell line. Methods With the technology of cell biology and molecular biology,we studied the phenomenon of growth inhibition of MDA MB 435 breast cancer cells by the novel retinoid,and the protein expression of cyclin D1 and cyclin E in the breast cancer cells treated with the drug. Results The growth of MDA MB 435 cells was significantly inhibited by AHPN.The protein expression of cyclin D1 in the breast cancer cells treated with AHPN was reduced with time course,but the expression of cyclin E did not change significantly. Conclusions The novel retinoid AHPN can effectively inhibit the growth of MDA MB 435 breast cancer cells.The growth inhibition may be induced by decreasing cyclin D1,and may has nothing to do with cyclin E.
