愈骨合剂促进骨折愈合作用的实验研究

摘    要:为研究愈骨合剂对骨折的治疗作用. 将36只日本大耳白兔,制成骨折动物模型,随机分为愈骨合剂组、接骨片组和生理盐水组,在第8、16、24、32天,分别进行X线观察、病理组织学检查、周围血细胞分析及血清酶学检测.结果显示X线观察在第16天愈骨合剂组的骨膜反应和骨痂生成均比对照组明显,全部有骨痂生成,其中7只有中等量骨痂生成.第24天,愈骨合剂组骨痂生成继续增多,其中2只骨折线开始模糊.第32天,愈骨合剂组有4只骨折线基本消失,骨痂密度与骨质接近.经Ridit检验,在第32天盐水组R=0.28,接骨片组R=0.52,愈骨合剂组R=0.70.愈骨合剂组与盐水组比较有显著性差异(P<0.05).病理组织学检查显示,骨折周围血肿等级( )及( ),盐水组共9例,R=0.5008,接骨片组共8例,R=0.4028,愈骨合剂组共9例,R=0.5880;骨痂等级( )及( ),盐水组共4例,R=0.3707,接骨片组共5例,R=0.4630,愈骨合剂组共7例,R=0.6574;软骨骨痂等级( )及( ),盐水组共2例,R=0.3380,接骨片组共5例,R=0.5972,愈骨合剂组共5例,R=0.5648;从R值分析愈骨合剂组比盐水组骨痂生长明显加快.血清酶学检验显示,盐水组用药前后ALP、CK及LDH的升高速度不显著(均为P>0.1),愈骨合剂组用药前后ALP、CK及LDH的升高速度显著(均为P<0.01).表明愈骨合剂具有明显促进骨痂形成的作用.

Abstract:In order to study the stimulative effect of "YUGU HEJI"(YGHJ,a TCD mixture for fracture healing) on fracture healing,36 Japanese white rabbits were made into animal models of fracture and randomly divided into three groups I, II and III,which were treated by YGHJ,by "JIEGU PIAN"(JGP,TCD tablets for fracture healing) and by normal saline,respectively;the X-ray observations,histopathological examinations,peripheral hemocytes analysis and seroenzymology assay were made on the three groups on 8th,16th,24th and 32nd days,respectively.The results included: a.X-ray Observations.On the 16th day,Group I had more periosteal reaction and more porosis than Group II,all rabbits had porosis,7 of them had moderate porosis;on the 24th day,the porosis in Group I continued to increase,among which the fracture line became obscure in 2 rabbits;on the 32nd day,the fracture line in 4 rabbits of Group I disappeared in the main,the callus density approached the bone substance;the Ridit test showed that on the 32nd day,R=0.28 in Group III,R=0.52 in Group II and R=0.70 in Group I,which had a significant difference from Group III (P < 0.05).b.Histopathological Examinations.The hematoma grades(+++) and(++++) around the fracture included R=0.5008 in 9 rabbits of Group III,R=0.4028 in 8 of Group II and R=0.5880 in 9 of Group I;the callus grades(+++) and(++++) included R=0.3707 in 4 rabbits of Group III,R=0.4630 in 5 of Group II and R=0.6574 in 7 of Group I;the cartilage callus grades(+++) and(++++) included R=0.3380 in 2 rabbits of Group III,R=0.5972 in 5 of Group II and R=0.5648 in 5 of Group I;the R-value analysis showed that the porosis of Group I was obviously speeded up in comparison with the one of Group III.c.Seroenzymology Assay.The ascending speeds of ALP,CK and LDH in Group III were not obvious (P > 0.1) before and after the treatment,while the ones in Group I were very significant(P < 0.01),suggesting that YGHJ has a significant function to promote porosis.
