芹龙颗粒抗急性肝损伤的实验研究

摘    要:目的观察芹龙颗粒(QL)对小鼠急性肝损伤的防治作用。方法将60只小鼠随机分为联苯双酯阳性药物组,正常动物组,CCl4模型对照组和QL大、中、小(10、5、2.5.gkg^-1)3个剂量组。药物组动物先分别灌胃给药3d,第4d,按体重腹腔注射0.5%浓度CCl4橄榄油溶液(10ml.kg^-1),复制急性肝损伤动物模型。24h后取血测定血清中ALT,AST含量;并取肝脏,称重,10%甲醛固定,HE染色,于光镜下观察肝脏组织学变化。结果模型组小鼠肝功能明显异常,肝组织变性、坏死严重;QL组能明显改善CCl4中毒小鼠肝功能,其降酶率(ALT)以大、中、小剂量分别为54.3%、49.1%、41.2%。肝组织学改变较轻,体重减少较模型组轻。结论QL对CCl4致小鼠急性肝损伤具有明显的保护作用,其改善肝功能具有良好的量效关系。

Abstract:Aim To investigate the effect of of Qin Long Granule (QL) on acute hepatic injury in mice. Methods Sixty mice were randomly divided into the bifendate control group, normal group, CCL4 model group and three QL groups (high (10g·kg^-1) ,middle (5g·kg^-1) and low (2.5g·kg^-1) dosage group). QL groups were administrated with QL by ig. for 3days. On the fouth day, the mice's acute hepatic injury model was induced by the intraperitoneal injection of 0.5% CCl4 ( 10ml ·kg^-1 ) and the blood sample was collected after 24h. Then the level of ALT and AST in serum was determined and histopathological changes were examined by optical microscopy. Results In the model group the level of ALT,AST increased significantly, and hepatic tissue degeneration was seen obviously. In QL groups, the level of ALT dropped significantly (the restrain rate of the high ,middle and low dosage group was 54.3% ,49.1% and 41.2% ,respectively) and hepatic tissue degeneration was less severe. Conclusion QL possesses protective effect on acute hepatic injury induced by CCl4. Its protective effect on the liver has a good dose-effect relationship.
