辛伐他汀对高糖诱导的肾小管上皮细胞转分化的影响

摘    要:目的:观察辛伐他汀对高糖诱导的肾小管上皮细胞转分化的作用。方法:大鼠肾小管上皮细胞随机分成三组(n=8):对照组,高糖组和辛伐他汀组。对照组加培养液,高糖和辛伐他汀组在培养液中加葡萄糖,终浓度为30mmol/L,辛伐他汀组加入终浓度为1μmol/L辛伐他汀。在6,12,24,48,72 h时用免疫细胞化学和Western blot检测α-SMA的表达。结果:对照组各时间段均无α-SMA表达。高糖组6 h时出现α-SMA阳性表达,48 h达高峰。与高糖组比较,辛伐他汀组各时间点α-SMA的表达均明显减少(P<0.05)。结论:辛伐他汀可抑制高糖诱导的大鼠肾小管上皮细胞转分化。

Abstract:Objective To investigate the effects of simvastatin in renal tubular epithelial cells transdifferentiation induced with high glucose.Methods Cultured renal tubular epithelial cells were randomly divided into three groups( n = 8 ) :control group (group C ) , high glucose group (group D) and simvastatin group( group S) , and given culture solution, high glucose (terminal concentration :30 mmoL/L), high glucose ( terminal concentration : 30 mmoL/L) plus simvastatin ( 1 μmoL/L), respectively. The expression of α-SMA was detected with immuocytochemistry and Western blot at 6,12,24,48 and 72 hours, respectively. Results The expression of α-SMA was not detected at all the time points. The positive expression of α-SMA in group D was detected at 6 h and reached peak at 48 h. The expression of α-SMA in group S was significantly lower than that of group D at all the time points( P 〈 0.05). Conclusion Simvastatin could inhibit the transdifferentiation of renal tubular epithelial cells induced with high glucose.
