苏拉明对体外培养的人卵巢癌细胞HO-8910PM增殖及乙酰肝素酶表达的影响

摘    要:目的探讨苏拉明对体外培养的人卵巢癌细胞增殖和乙酰肝素酶基因表达的影响。方法不同浓度苏拉明作用于人卵巢癌细胞株HO-8910PM,用四甲基偶氮唑蓝(MTT)比色法检测HO-8910PM增殖情况;免疫细胞化学方法和原位杂交方法检测苏拉明作用前后HO-8910PM乙酰肝素酶基因mRNA及蛋白表达变化。结果MTT结果显示50、100、200、300、400、500、600μg/ml浓度的苏拉明作用于HO-8910PM细胞96h的生长抑制率(GI)分别是13.1%、22.6%、35.1%、43.7%、54.3%、64.8%、70.9%(P0.01),不同时间及不同浓度组吸光光度值A差异有统计学意义(P0.01)。对照组比较苏拉明作用48h后,HO-8910PM细胞乙酰肝素酶基因mRNA及蛋白表达显著降低(P0.01)。结论苏拉明以时间及剂量效应方式抑制体外培养的HO-8910PM增殖;同时下调乙酰肝素酶基因mRNA及蛋白表达;提示苏拉明作为HPA抑制剂有望成为转移性卵巢癌联合化疗用药。

Abstract:Objective To observe the effect of suramin on the proliferation of cultured human ovarian cancer cells and it's effect on the expression of heparanase mRNA.Methods Human ovarian cancer cell line HO-8910PM were treated with different concentration of suramin (50,100,200,300,400,500,600 μg/ ml), cell proliferation was measured by MTT. The expression of heparanase (HPA) mRNA and protein were detected by in-suit hybridization and immunocytochemistry.Results The growth inhibition ratios in each suramin treatment group on the 4th day were 13.1%, 22.6%, 35.1%, 43.7%, 54.3%, 64.8% and 70.9%, respectively, there were significantly difference between each group (P〈0.01). The expression of heparanase mRNA and protein in HO-8910PM cells were significantly higher than that in HO-8910PM cells treated with suramin.Conclusion Suramin could inhibit the proliferation of HO-8910PM cells with time-dependent and dose-dependent manner, also it can down-regulate the expression of HPA in HO-8910PM cells.
