基于SELDI-TOF-MS技术的慢性胃炎患者腻苔相关蛋白标志物研究

目的 研究慢性胃炎患者腻苔相关蛋白标志物.方法 采集慢性胃炎患者的舌苔100例、正常人的舌苔38例,根据舌苔色质将慢性胃炎患者分为腻苔组50例,非腻苔组50例,其中腻苔组又分为白腻苔组17例,黄腻苔组33例.采用表面增强激光解析离子化飞行时间质谱技术对各组舌苔进行蛋白质组学研究.利用NP20蛋白芯片技术进行舌苔蛋白质组学研究,用蛋白芯片阅读器PBSII对芯片进行扫描、分析.结果 在质荷比1000～20000范围内,腻苔组与正常对照组有69个差异蛋白峰(P＜0.01),非腻苔组与正常对照组有63个差异蛋白峰(P＜0.01),腻苔组与非腻苔组有4个差异蛋白峰(P＜0.01),白腻苔组与正常对照组有52个差异蛋白峰(P＜0.01),黄腻苔组与正常对照组有62个差异蛋白峰(P＜0.01),白腻苔组与黄腻苔组有2个差异蛋白峰(P＜0.01).结论 该研究初步筛选出了慢性胃炎患者腻苔的蛋白标志物,为慢性胃炎中 医舌诊的客观化研究提供了一定的实验依据.

Research on Associated Protein Markers in Greasy Tongue Coating of Chronic Gastritis Patients with SELDI-TOF-MS Technique

Objective To investigate the associated protein markers in tongue coating of chronic gastritis patients.Methods The tongue coating was taken from 100 chronic gastritis patients and 38 healthy persons.The patients were divided to the greasy tongue coating group(50 cases) and non-greasy tongue coating group(50 cases also),and the greasy tongue coating group were further grouped into the white-greasy tongue coating group(17 cases) and the yellow-greasy tongue coating group(33 cases).Surface-enhanced laser desorption/ionization time of flight mass spectrometry(SELDI-TOF-MS) technique was applied for proteomic study on the tongue coatings.Results Within the range of 1000 to 20000 M/Z,there were 69 different protein peaks identified between the greasy tongue coating group and the control group(P<0.01).Between the non-greasy tongue coating group and the control group,63 different protein peaks were identified(P<0.01).Between the greasy tongue coating group and non-greasy tongue coating group,4 different protein peaks were identified(P<0.01).Between the white-greasy tongue coating group and the control group,52 different protein peaks were identified(P<0.01).Between the yellow-greasy tongue coating group and the control group,62 different protein peaks were identified(P<0.01).Between the white-greasy tongue coating group and yellow-greasy tongue coating group,2 different protein peaks were identified(P<0.01).Conclusion The research screens the protein markers in greasy tongue coating of chronic gastritis patients and provides experimental evidence for the tongue diagnosis study of chronic gastritis.
