克瘤丸对结肠癌细胞HT-29细胞凋亡及细胞周期的影响

摘    要:目的 观察克瘤丸对人结肠癌细胞生长的影响及其作用机制.方法 将4mg/ml、2mg/ml、1mg/ml、0.5mg/ml、0.25mg/ml浓度克瘤丸作用于HT-29细胞24~48h后,用四甲基偶氮唑蓝(MTT)法检测克瘤丸对HT-29细胞增殖的影响,AnnexinV单染法检测克瘤丸对HT-29细胞凋亡的影响,流式细胞仪检测细胞周期的分布情况. 结果 与对照组比较,浓度为4、2、1、0.5mg/ml克瘤丸作用HT-29细胞48h,OD值、48h抑制率差异均有统计学意义(P<0.01);4、2、1mg/ml浓度克瘤丸作用HT-29细胞48h,OD值、48h抑制率均明显优于0.25mg/ml及0.5mg/ml浓度克瘤丸(P<0.01).与1mg/ml浓度比较,2mg/ml、4mg/ml组凋亡率差异有统计学意义(P<0.05).4mg/ml时G0/G1期前出现明显的亚二倍体区,与1mg/ml、2mg/ml组比较,差异有统计学意义(P<0.05).结论 克瘤丸对人结肠癌细胞HT-29的增殖具有抑制作用并能够引起该细胞的凋亡.

Abstract:Objective To observe the effect and mechanism of Keliu Pill ( antitumor pill ) on the growth of human colon car- cinoma cell.Methods The cell proliferation was assessed by methyl-thiazolyl-tetrazolium ( MTT ) assay after 24to 48hours of treatment with different concentrations ( 4 , 2 , 1 , 0.5or 0.25mg / ml ) of Keliu Pill on HT-29cells.The apoptosis was deter- mined with AnnexinⅤstaining and cell cycleand was determined with flow cytometry ( FCM ) .Results Comparing with the control group , Keliu Pill at different concentrations ( 0.5to 4mg / ml ) could significantly inhibit the proliferation of HT-29 cells , with statistical significance in the 48hcellular proliferation inhibition rate ( CPIR ) and optical density ( OD ) value ( P< 0.01 ) .The OD values and 48hCPIRs of Keliu Pill at high concentrations ( 1mg / ml to 4mg / ml ) were superior to those at low concentrations ( 0.25mg / ml and 0.5mg / ml ), with statistical significance ( P<0.01 ) .Comparing with the group of 1mg / ml , the apoptosis rates were significantly increased in group of 2and 4mg / ml of Keliu Pill ( P<0.05 ) .The apparent hypodiploid area appeared before G0 / G1phase after treatment with 4mg / ml of Keliu Pill , with significant difference as compared with 1mg / ml and 2mg / ml of Keliu Pill ( P<0.05 ) .Conclusion Keliu Pill could inhibit the proliferation and induce the apoptosis of hu- man colon carcinoma cell line HT-29.
