S100B与NSE水平变化在大鼠脑室出血后脑损伤中的研究

摘    要:目的探讨大鼠脑室出血(IVH)损伤时脑脊液中S100B蛋白与神经元特异性醇化酶(NSE)水平的变化。判断不同血肿IVH后脑损伤的严重程度。方法选用SD雄性大鼠采用随即数字法分成3组:正常组,假手术组,IVH组,IVH组又分为15ul和30ul组。采用脑立体定向自体血注入法做成IVH模型,1天后采集大鼠脑脊液(CSF),酶联免疫吸附法检测S100B和NSE的水平,电镜观察脑细胞超微结构改变。结果IVH后1天S100B与NSE均在IVH组较正常与假手术组均显著升高(P〈0.01)。15ul与30ul两组间S100B水平变化比较有显著差异(P〈0.05),NSE在15ul与30ul两组间则无统计学差异(P〉0.05)。超微结构主要表现为线粒体肿胀,嵴减少或消失,粗面内质网脱颗粒,细胞质水肿等,15ul相对30ul组脑细胞改变较轻。结论NSE与S100B能够较敏感的作为IVH后的脑损伤的标志物,而S100则对不同血肿量所致的IVH后脑损伤更为敏感。

Abstract:Objective To investigate the levels of protein S100B and neuron - specific enolase (NSE) in intraventricular hemorrhage (IVH) in rats, and to judge the extent of brain damage after IVH in different blood volumes. Method The rats divided into three groups : normal group ( N), sham group (S) and the IVH group ( M), which is subdivided into two group: 15 ul group ( M 1 ) and 30ul group(M2). A model of IVH was established by stereotactically injecting the autologous femoral artery blood into the right lateral ven- tricle in rats. The CSF was obtained from rats after 24hours. The levels of S100B and NSE were measured by enzyme linked immu- nosorbent assay. The ultrastructure changes of neurons were also observed. Results The increased S100B and NSE levels in M1 and M2 groups was significantly compared with N and S groups(P 〈0.01 ). For S100B concentrations, there was a significant differences between M1 group and M2 group(P 〈0.05). No other significant differences were found between groups. The important ultrastructure changes were that mitochondria swelling, cristae reduction or disappearance, rough endoplasmic reticulam degranulation, cytoplasm oedema. The ultrastructure changes in MI group were comparatively gentle than M2 group. Conclusion Our results suggest that S100B and NSE could be a sensitive marker of brain damage of intraventricular hemorrhage, whereas S100B is more sensitive for different hematoma volumes of IVH in rats.
